      Penyusunan Draf Genom Mitokondria Badak Sumatera (Dicerorhinus sumatrensis) Tingkat Kelengkapan Tinggi dengan PromethION 2 Solo

      Abstract
      Badak sumatera (Dicerorhinus sumatrensis) merupakan salah satu mamalia yang masuk dalam kategori Critically Endangered oleh IUCN. Upaya pelestariannya sangat bergantung pada ketersediaan informasi genetik yang mendalam. Penelitian ini bertujuan menyusun draft genom mitokondria dari individu badak sumatera bernama Ratu yang berada di Suaka Rhino Sumatera (SRS) Way Kambas. Teknologi long-read sequencing menggunakan PromethION 2 Solo (Oxford Nanopore Technologies) berhasil merakit whole genome sebesar 2,55 Gb dengan kualitas rata-rata Q15,9. Perakitan genom mitokondria menggunakan GetOrganelle menghasilkan contig sirkular sepanjang 16.739 bp. Anotasi mitokondria dilakukan menggunakan MitoAnnotator, dilanjutkan dengan analisis filogenetik berdasarkan gen COX1. Hasil menunjukkan bahwa individu Ratu membentuk clade monofiletik bersama kelompok D. sumatrensis lainnya dengan dukungan bootstrap sebesar 100%. Temuan ini menjadi dasar penting dalam pemetaan genetik spesies langka ini dan mendukung strategi konservasi berbasis genomik.
       
      The sumatran rhinoceros (Dicerorhinus sumatrensis) is among the world’s most critically endangered mammals, as classified by the IUCN. Effective conservation of this species depends greatly on the availability of detailed and high quality genetic information. This study aimed to assemble the mitochondrial draft genome of an individual Sumatran rhinoceros named "Ratu" from the Sumatran Rhino Sanctuary (SRS) in Way Kambas. Long-read sequencing technology using PromethION 2 Solo (Oxford Nanopore Technologies) successfully generated a whole-genome dataset of 2.55 Gb with an average read quality of Q15.9. Mitochondrial genome assembly was carried out using GetOrganelle, resulting in a circular contig of 16,739 bp. Annotation was performed using MitoAnnotator, followed by phylogenetic analysis based on the COX1 gene. The results showed that Ratu forms a monophyletic clade with other D. sumatrensis sequences (100% bootstrap). These findings provide crucial genetic insights for this endangered species and support genome-based conservation strategies.
