      Pengaruh Konsentrasi Silika dan Media Tanam terhadap Keberhasilan Aklimatisasi dan Pertumbuhan Planlet Dendrobium violaceoflavens x Dendrobium canaliculatum

      Abstract
      Salah satu jenis Anggrek Dendrobium section Spatulata hasil hibridisasi yang baru dikembangkan di Indonesia adalah jenis Dendrobium violaceoflavens x Dendrobium canaliculatum yang merupakan hasil silangan dari Dendrobium violaceoflavens dan Dendrobium canaliculatum. Perbanyakan melalui metode kultur jaringan dapat menjadi salah satu alternatif perbanyakannya, namun metode ini memiliki fase aklimatisasi yang sangat rentan. Penelitian ini bertujuan mengetahui pengaruh konsentrasi silika dan jenis media tanam pada pertumbuhan Dendrobium violaceoflavens x Dendrobium canaliculatum pada masa aklimatisasi. Rancangan yang digunakan adalah rancangan kelompok lengkap teracak (RKLT) faktor split plot dengan dua faktor. Faktor pertama adalah konsentrasi silika (SiO2) dengan empat taraf konsentrasi, yaitu 0 g l-1, 0,01 g l-1, 0,02 g l-1 dan 0,03 g l-1. Faktor kedua yaitu perlakuan media tanam sebanyak tiga jenis yaitu spaghnum moss, moss hitam dan serabut kelapa. Hasil penelitian menunjukkan perlakuan konsentrasi silika pada planlet Dendrobium violaceoflavens x Dendrobium canaliculatum tidak memberikan pengaruh nyata terhadap jumlah daun, tinggi tanaman, kerapatan stomata, ketebalan epidermis dan persentase hidup. Sementara itu, perlakuan berbagai media tanam berpengaruh nyata pada jumlah daun, tinggi tanaman, kerapatan stomata, ketebalan epidermis dan persentase hidup. Media tanam terbaik adalah moss hitam dan serabut kelapa. Interaksi perlakuan konsentrasi silika dan media tanam berpengaruh sangat nyata pada ketebalan epidermis dan berpengaruh nyata pada kerapatan stomata.
       
      One of the newly developed hybrids of Dendrobium orchids from the Spatulata section in Indonesia is Dendrobium violaceoflavens × Dendrobium canaliculatum, a cross between Dendrobium violaceoflavens and Dendrobium canaliculatum. Micropropagation through tissue culture is a potential method for large-scale propagation; however, this technique involves a highly vulnerable acclimatization phase. This study aimed to investigate the effects of silica concentration and growing media types on the growth of Dendrobium violaceoflavens × Dendrobium canaliculatum during the acclimatization period. The experimental design used was a split-plot randomized complete block design (RCBD) with two factors. The first factor was silica (SiO2) concentration at four levels: 0 g l?¹, 0,01 g l?¹, 0,02 g l?¹, and 0,03 g l?¹. The second factor was the growing media, consisting of three types; sphagnum moss, black moss, and coconut fiber. The results showed that silica concentration had no significant effect on leaf number, plant height, stomatal density, epidermal thickness, or survival rate. In contrast, growing media significantly affected all observed parameters. The best-performing media were black moss and coconut husk. Additionally, a highly significant interaction between silica concentration and growing media was observed for epidermal thickness, and a significant interaction was found for stomatal density.
