      Efektivitas Ekstrak Daun Rimbang (Solanum Torvum) terhadap Penyembuhan Luka Kulit Tikus Sprague Dawley

      Abstract
      Penyembuhan luka tetap menjadi fokus penting dalam penelitian klinis dan eksperimental, dengan meningkatnya minat pada biomaterial sintetis dan alami untuk aplikasi terapeutik. Intervensi berbasis herbal telah menarik perhatian karena potensinya dalam mendorong perbaikan jaringan. Studi ini mengevaluasi efikasi penyembuhan luka gel ekstrak daun S. torvum (rimbang) melalui pemodelan in silico, penilaian makroskopis dan analisis histopatologi dalam model kulit tikus. Hasil uji determinasi tumbuhan mengkonfirmasi bahwa tumbuhan yang digunakan pada penelitian ini adalah tumbuhan rimbang, jenis Solanum torvum. Hasil uji fitokimia menunjukkan bahwa ekstrak etanol daun rimbang mengandung alkaloid, flavonoid, steroid dan terpenoid, saponin, serta tanin. Tikus dibagi menjadi lima kelompok yang menerima terapi berbeda: gel ekstrak 5% (EDR 5%), ekstrak 20% (EDR 20%), ekstrak 35% (EDR 35%), Bioplacenton®/Neomycin sebagai kontrol positif (KP) dan tanpa terapi sebagai kontrol negatif (KN). Parameter penyembuhan luka punch biopsy kulit dinilai pada hari ke- 3, 5 dan 7 tanpa dan dengan terapi, termasuk pengukuran luas area luka, infiltrasi sel radang, angiogenesis, re- epitelisasi, dan kepadatan kolagen. Analisis in silico menunjukkan potensi aktivitas antimikroba ekstrak melalui penghambatan reseptor dihidrofolat reduktase (DHFR), didukung oleh nilai ?G dan Ki yang positif. Evaluasi makroskopis dan mikroskopis menunjukkan bahwa terapi topikal ekstrak daun rimbang, terutama pada konsentrasi 35%, secara signifikan mempercepat penyembuhan luka kulit, hal ini dibuktikan dengan berkurangnya infiltrasi sel radang, peningkatan angiogenesis, peningkatan re-epitelisasi, dan kepadatan kolagen yang lebih baik. Temuan ini mendukung potensi terapeutik ekstrak daun rimbang sebagai agen alami untuk penanganan luka kulit secara topikal.
       
      Wound healing remained an important focus in both clinical and experimental research, with growing interest in synthetic and natural biomaterials for therapeutic applications. Herbal-based interventions had attracted attention due to their potential in promoting tissue repair. This study evaluated the wound- healing efficacy of S. torvum (rimbang) leaf extract gel through in silico modeling, macroscopic assessment, and histopathological analysis in a rat skin model. Plant determination tests confirmed that the plant used in this study was Solanum torvum. Phytochemical screening revealed that the ethanol extract of rimbang leaves contained alkaloids, flavonoids, steroids and terpenoids, saponins, as well as tannins. Rats were divided into five groups receiving different treatments: 5% extract gel (EDR 5%), 20% extract gel (EDR 20%), 35% extract gel (EDR 35%), Bioplacenton®/Neomycin as the positive control (PC), and no treatment as the negative control (NC). Parameters of skin punch biopsy wound healing were assessed on days 3, 5, and 7 with and without treatment, including wound area measurement, inflammatory cell infiltration, angiogenesis, re-epithelialization, and collagen density. In silico analysis showed the antimicrobial potential of the extract through inhibition of the dihydrofolate reductase (DHFR) receptor, supported by favorable ?G and Ki values. Macroscopic and microscopic evaluations demonstrated that topical treatment with rimbang leaf extract, particularly at a concentration of 35%, significantly accelerated skin wound healing, as evidenced by reduced inflammatory cell infiltration, increased angiogenesis, improved re- epithelialization, and higher collagen density. These findings supported the therapeutic potential of rimbang leaf extract as a natural topical agent for skin wound management.
