      Aktivitas Antibakteri dan Sitotoksisitas dari Kombucha Temu Putih (Curcuma zedoaria)

      Abstract
      Temu putih (Curcuma zedoaria) mengandung senyawa bioaktif yang berpotensi sebagai antibakteri dan bersifat sitotoksik, namun kandungan senyawa dan bioaktivitasnya masih tergolong rendah. Sehingga, fermentasi diperlukan untuk meningkatkan potensinya. Penelitian ini bertujuan mengevaluasi aktivitas antibakteri dan sitotoksisitas rimpang temu putih yang difermentasi menggunakan Symbiotic Culture of Bacteria and Yeast (SCOBY). Uji antibakteri terhadap Escherichia coli dan Staphylococcus aureus dilakukan dengan metode difusi cakram, Konsentrasi Hambat Minimum (KHM), dan Konsentrasi Bunuh Minimum (KBM), sementara untuk analisis sifat sitotoksik digunakan Brine Shrimp Lethality Test (BSLT). Hasil menunjukkan bahwa fermentasi meningkatkan efektivitas antibakteri jika dibandingkan dengan sampel non-fermentasi, dengan zona hambat 13,7 ± 0,7 mm untuk E. coli dan 12,8 ± 0,4 mm untuk S. aureus. Aktivitas sitotoksik juga meningkat, ditunjukkan oleh nilai LC50 kombucha sebesar 145,292 µg/mL (toksik sedang) dibandingkan non-fermentasi sebesar 1381,793 µg/mL (tidak toksik). Dengan demikian, fermentasi terbukti dapat meningkatkan aktivitas antibakteri dan sitotoksisitasnya secara signifikan.
       
      White turmeric (Curcuma zedoaria) contains bioactive compounds with potential antibacterial and cytotoxic properties, however, the quantity of these compounds and their biological activity remain relatively low. Therefore, fermentation is required to enhance their potency. This study evaluates the antibacterial and cytotoxic activity of C. zedoaria rhizome fermented using a Symbiotic Culture of Bacteria and Yeast (SCOBY). Antibacterial activity against Escherichia coli and Staphylococcus aureus was assessed using the disk diffusion method, Minimum Inhibitory Concentration (MIC), and Minimum Bactericidal Concentration (MBC), while cytotoxic activity was analyzed using the Brine Shrimp Lethality Test (BSLT). The results showed that fermentation enhanced antibacterial effectiveness compared to the non-fermented samples, with inhibition zones of 13.7 ± 0.7 mm for E. coli and 12.8 ± 0.4 mm for S. aureus. Cytotoxic activity also increased, indicated by an LC50 value of 145.292 µg/mL for the kombucha (moderately toxic), compared to 1381.793 µg/mL for the non-fermented sample (non-toxic). Thus, fermentation significantly enhances its antibacterial and cytotoxic activities.
