Gandarusa (Justicia gendarussa) is a medicinal plant that contains natural antioxidant compounds. Fermentation using Aspergillus oryzae has the potential to enhance the plant’s antioxidant activity. This study aimed to examine and compare the antioxidant activity of fractions from column chromatography of gandarusa extract, between non-fermented and fermented samples. Gandarusa was fermented for 14 days, then extracted using methanol as the solvent. The extract was subsequently purified using column chromatography with varying ratios of ethyl acetate and methanol as eluents. The resulting extracts were analyzed for total phenolic content, quantitative antioxidant activity using the DPPH method, and qualitative antioxidant activity using the TLC bioautography method. The results showed that the fraction eluted with ethyl acetate:methanol in a 6:4 ratio, both in non-fermented and fermented samples, yielded the highest total phenolic content and antioxidant activity. However, there was no significant difference in total phenolic content or antioxidant activity between the fractions of non-fermented and fermented gandarusa extracts.
