      Konservasi In Vitro Kantong Semar (Nepenthes rafflesiana Jack.)

      Abstract
      Nepenthes rafflesiana merupakan tumbuhan karnivora yang memiliki organ berbentuk kantong untuk menangkap mangsa. N. rafflesiana memiliki manfaat sebagai tanaman hias sekaligus tanaman obat, namun jumlahnya terus menurun di alam. Penelitian ini bertujuan mengidentifikasi pengaruh pemberian hormon thidiazuron (TDZ) pada pertumbuhan eksplan N. rafflesiana secara kultur jaringan. Terdapat empat jenis konsentrasi thidiazuron yang digunakan yaitu 0 mg/L, 0,01 mg/L, 0,1 mg/L, dan 1 mg/L. Pengamatan eksplan dilaksanakan selama 10 minggu setelah tanam, di mana parameter yang diamati mencakup jumlah daun, panjang daun, dan tinggi tanaman. Hasil penelitian menunjukkan konsentrasi TDZ 0,1 mg/L memberikan hasil terbaik pada pertumbuhan jumlah daun, sedangkan konsentrasi TDZ 0 mg/L memberikan hasil terbaik pada pertumbuhan panjang daun dan tinggi tanaman. Hasil analisis menunjukkan bahwa hormon thidiazuron memiliki pengaruh beda nyata pada parameter panjang daun dan tinggi tanaman, sedangkan pada parameter jumlah daun tidak memiliki pengaruh beda nyata.
       
      Nepenthes rafflesiana is a carnivorous plant that has pitcher-shaped organs to capture prey. N. rafflesiana has benefits as both an ornamental and medicinal plant, but its population continues to decline in the wild. This study aimed to identify the effect of thidiazuron (TDZ) hormone on the growth of N. rafflesiana explants in tissue culture. There were four concentrations of thidiazuron used, namely 0 mg/L, 0.01 mg/L, 0.1 mg/L, and 1 mg/L. Observations of explants were carried out for 10 weeks after planting, where the parameters observed included the number of leaves, leaf length, and plant height. The results showed that 0.1 mg/L TDZ gave the best results for leaf number growth, while 0 mg/L TDZ gave the best results for leaf length and plant height growth. The analysis showed that thidiazuron hormone had a significant effect on the parameters of leaf length and plant height, but no significant effect on the parameter of leaf number.
