      Aplikasi Sitokinin BAP dan Auksin NAA untuk Multiplikasi Tunas Pisang Kepok dan Ambon secara In vitro

      Abstract
      Pisang merupakan komoditas yang banyak diminati oleh konsumen di dalam maupun di luar negeri, dibuktikan dengan meningkatnya angka konsumsi dan produksi pisang di dunia. Peningkatan produksi pisang menghadapi keterbatasan bahan tanam yang berkualitas, sehingga diperlukan peningkatan produksi bibit menggunakan kultur in vitro. Penelitian bertujuan mengetahui respon pertumbuhan pisang kepok dan ambon secara in vitro serta mendapatkan kombinasi konsentrasi BAP dan NAA yang optimal untuk multiplikasi tunas pisang kepok dan ambon secara in vitro. Penelitian dilaksanakan pada bulan Maret hingga Juni 2025 di Laboratorium Kultur Jaringan I, IPB University. Penelitian menggunakan Rancangan Kelompok Lengkap Teracak (RKLT) dengan dua faktor yaitu jenis pisang (Kepok dan Ambon), dan kombinasi konsentrasi BAP dan NAA. Kombinasi BAP (4 dan 6 mg L-1) dan NAA (0,5 dan 1 mg L-1) tidak mampu menginduksi pembentukan tunas baru pada eksplan pisang ambon dan kepok selama masa pengamatan. Perbedaan jenis pisang mempengaruhi waktu munculnya akar dan jumlah daun pada 10 MSP dan 12 MSP, tinggi tanaman dan diameter batang. Pisang kepok memiliki respon waktu muncul akar yang lebih cepat, jumlah daun yang lebih banyak serta tanaman yang lebih tinggi dan diameter yang lebih besar dibandingkan pisang ambon.
       
      Banana (Musa sp.) is one of the most popular comodity among global consumers, as evidenced by the increase in its consumption and production worldwide. However, banana production faces a major constraint: the limited availability of high-quality planting materials. In vitro culture techniques are needed to enhance seedling production. This study aimed to determine the optimal combination of BAP and NAA concentrations for shoot multiplication. The experiment was conducted from March to June 2025 at the Tissue Culture Laboratory I, IPB University. This study used randomized complete block design (CRBD) with two factors: banana cultivars (Kepok and Ambon) and combinations of BAP and NAA concentrations. Results showed that the combination of BAP (4 and 6 mg L?¹) and NAA (0,5 and 1 mg L?¹) failed to induce the formation of new shoots on Ambon and Kepok banana explants during the observation period. The banana genotype influenced the time of root and leaf emergence at 10 and 12 weeks after planting (WAP), as well as plant height and stem diameter. Kepok showed a faster response in root emergence, higher number of leaves with greater plant height and stem diameter compared to ambon.
