      Isolasi dan Karakterisasi Bacillus spp. Penghasil Enzim Hidrolitik dari Saluran Pencernaan Ikan Bawal Air Tawar (Colossoma macropomum)

      Abstract
      Tingginya permintaan ikan bawal air tawar sebagai ikan konsumsi dan biaya pakan yang tinggi dalam akuakultur dapat memberikan tekanan yang signifikan bagi para pembudidaya ikan bawal air tawar di Indonesia. Pendekatan mikroba dalam hal ini diperlukan sebagai solusi yang hemat biaya dan berkelanjutan terhadap lingkungan. Penelitian ini bertujuan mengisolasi dan menyeleksi Bacillus spp. berdasarkan aktivitas amilolitik, proteolitik, dan lipolitik dari saluran pencernaan ikan bawal air tawar (Colossoma macropomum). Sebanyak empat belas isolat diperoleh menggunakan metode renjatan panas. Semua isolat menunjukkan genus Bacillus spp., yakni basil Gram positif dan menghasilkan endospora. Uji hemolisis menunjukkan bahwa tujuh isolat memiliki aktivitas hemolitik gamma pada agar darah domba. Sebanyak tujuh isolat dilanjutkan pengujian aktivitas enzimatik, berupa amilolitik, proteolitik, dan lipolitik yang dievaluasi masingmasing pada 1% starch agar, 2% skim milk agar, dan 2% Tween 80 agar. Isolat BBW 4 dan BBW 5 menunjukkan ketiga aktivitas hidrolitik. Indeks amilolitik tertinggi diperoleh BBW 9 sebesar 2,15; indeks proteolitik oleh BBW 14 sebesar 2,09; dan indeks lipolitik oleh BBW 7 sebesar 1,24. Isolat-isolat tersebut diduga berpotensi meningkatkan produktivitas dan efisiensi pakan untuk budidaya ikan bawal air tawar.
       
      The high demand for tambaqui as a consumption fish and the high feed costs in aquaculture have potentially exerted considerable pressure on tambaqui farmers in Indonesia. A microbial approach is needed as a cost-effective and environmentally sustainable solution. This study aims to isolate and select Bacillus spp. based on amylolytic, proteolytic, and lipolytic activity from the digestive tract of tambaqui (Colossoma macropomum). The heat shock method was used to obtain a total of fourteen isolates. All of the isolates indicated the Bacillus spp. strains due to positive Gram bacilli and endospore formation. The hemolytic assay showed that seven isolates had gamma hemolytic activity on sheep blood agar. The seven isolates were assessed for amylolytic, proteolytic, and lipolytic activity qualitatively on 1% starch agar, 2% skim milk agar, and 2% Tween 80 agar, respectively. The highest amylolytic index was obtained by BBW 9 at 2.15; proteolytic index by BBW 14 at 2.09; and lipolytic index by BBW 7 at 1.24. The isolates of BBW 4 and BBW 5 show all three hydrolytic activities. The isolate has the potential to enhance productivity and feed efficiency for tambaqui farming.
