      Induksi Ovulasi dengan Pemberian Rekombinan Luteinizing Hormone (LH) pada ikan lele Clarias gariepinus

      Abstract
      Ketersediaan hormon reproduksi buatan yang efektif dan berkelanjutan menjadi salah satu faktor penting dalam mendukung reproduksi ikan budidaya secara terkontrol. Teknologi rekombinan menjadi salah satu alternatif untuk memproduksi hormon secara efisien dan spesifik. Penelitian ini bertujuan untuk mengevaluasi hormon rekombinan luteinizing hormone ikan gurami (rLHg) terhadap induksi ovulasi pada ikan lele betina (Clarias gariepinus). Bakteri Escherichia coli BL21 (DE3) yang membawa vektor ekspresi rLHg, dikonfirmasi menggunakan PCR, dan konsentrasi protein rLHg diukur dengan metode Bradford. Rancangan percobaan yang digunakan adalah rancangan acak lengkap dengan tiga perlakuan dan empat ulangan, yaitu kontrol (K) berupa injeksi bufer fosfat salin (PBS), R1 (rLHg 0,1 mg/kg), dan R5 (rLHg 0,5 mg/kg bobot ikan). Induksi dilakukan satu kali secara intramuskular. Parameter yang diamati meliputi tingkat kelangsungan hidup, tingkat ovulasi, masa laten ovulasi, jumlah telur yang diovulasikan, diameter telur, dan pergerakan inti telur. Hasil penelitian menunjukkan bahwa perlakuan R5 mampu menginduksi ovulasi dengan tingkat keberhasilan 50%, sedangkan perlakuan R1 dan kontrol tidak menunjukkan ovulasi. Perlakuan R5 juga menghasilkan diameter telur yang lebih besar dan persentase telur fase germinal vesicle breakdown yang lebih tinggi. Seluruh ikan dari semua perlakuan menunjukkan tingkat kelangsungan hidup sebesar 100%. Berdasarkan hasil tersebut, dapat disimpulkan bahwa hormon rLHg pada dosis 0,5 mg/kg mampu menginduksi pematangan akhir dan ovulasi pada ikan lele betina.
       
      The application of artificial reproductive hormones is a crucial factor in supporting controlled breeding in aquaculture. Recombinant technology offers an efficient and targeted alternative for hormone production. This study aimed to evaluate the effectiveness of recombinant luteinizing hormone derived from giant gourami (rLHg) in inducing ovulation in female African catfish (Clarias gariepinus). Escherichia coli BL21 (DE3) strain competent cells carrying the rLHg expression vector, confirmed through PCR, and the concentration of recombinant protein was quantified using the Bradford assay. A completely randomized design was employed, consisting of three treatments with four replicates each: control (K, PBS injection), R1 (rLHg at 0,1 mg/kg fish body weight), and R5 (rLHg at 0,5 mg/kg fish body weight). Hormone induction was performed via a single intramuscular injection. Observed parameters included survival rate, ovulation rate, latency period, number of ovulated eggs, egg diameter, and germinal vesicle movement. The results showed that R5 treatment induced ovulation with a success rate of 50%, while no ovulation was observed in R1 and control groups. R5 also resulted in larger egg diameters and a higher proportion of oocytes at the germinal vesicle breakdown stage. All fish in every treatment exhibited a 100% survival rate. These findings indicate that rLHg at a dose of 0,5 mg/kg is capable of inducing final oocyte maturation and ovulation in female catfish.
