      Abstract

Antimicrobial peptides (AMPs) are natural compounds with promising potential as antibiotic alternatives to combat bacterial resistance. One notable AMP is snakin, known for its broad-spectrum activity and high cysteine content. This study aimed to isolate DNA and identify the snakin-encoding gene in Solanaceae plants commonly found in Indonesia, including black nightshade (leunca), potato, red chili, bird’s eye chili, and round eggplant. DNA isolation was carried out using the CTAB method, with additional treatment using liquid nitrogen, RNase, and Proteinase K. Analysis using a Nanodrop spectrophotometer (A260/A230 and A260/A280) and gel electrophoresis showed that these treatments produced DNA with suitable purity and concentration for PCR. PCR amplification was performed using SN1 primers, with various annealing temperatures to determine optimal conditions, and the results were visualized using agarose gel electrophoresis. The optimal annealing temperature was found to be 53?°C, producing a specific amplicon of 680 bp. The snakin gene was successfully identified in red chili and black nightshade, but not detected in potato, bird’s eye chili, or round eggplant. These findings suggest that the presence of the snakin gene varies among Solanaceae species. This study supports the potential development of AMPs from local plants as alternative antibacterial agents.
