      Penggunaan Serum Darah Domba sebagai Nutrisi dalam Bahan Pengencer Semen Domba Garut (Ovis aries)

      Abstract
      Domba Garut (Ovis aries) merupakan salah satu ternak ruminansia kecil yang banyak digemari masyarakat, akan tetapi reproduksinya lambat. Inseminasi buatan merupakan salah satu langkah yang efektif dalam menangani permasalahan tersebut. Inseminasi buatan dapat menggunakan media semen cair yang berisi kombinasi berbagai komponen, salah satunya serum darah. Penelitian ini bertujuan untuk mengetahui efektifitas serum darah dengan konsentrasi berbeda terhadap motilitas dan keutuhan membran plasma (MPU) spermatozoa domba Garut (Ovis aries) di sekolah vokasi IPB. Metode penelitian penelitian mencakup penampungan semen, pemeriksaan semen segar, pembuatan media pengencer, pengenceran semen, dan evaluasi semen cair. Perlakuan dikelompokkan menjadi: P1: Tris buffer + Soya Lesitin 1% + Serum darah 0% (without serum), P2: Tris buffer + Soya Lesitin 1% + Serum darah 5%, P3: Tris buffer + Soya Lesitin 1% + Serum darah 10% Hasil penelitian menunjukkan adanya perbedaan signifikan (P<0,05) pada parameter motilitas spermatozoa dengan hasil terbaik ditunjukkan oleh perlakuan 3 dengan konsentrasi serum darah 10%. Hasil pemeriksaan parameter keutuhan membran plasma (MPU) spermatozoa tidak menunjukkan adanya perbedaan signifikan (P>0,05).
       
      Garut sheep (Ovis aries) is one of the small ruminant livestock that is widely favored by the community, but its reproduction is slow. Artificial insemination is one of the effective steps in dealing with this problem A liquid semen medium for artificial insemination can be made by combining a number of ingredients, including blood serum. The purpose of this study is to ascertain how well blood serum at various levels affects the motility and plasma membrane integrity (MPU) of Garut Sheep spermatozoa at IPB Vocational School. The research methods include semen collection, fresh semen examination, dilution media preparation, semen dilution, and evaluation of liquid semen. The treatments were grouped into: P1: Tris buffer + Soya Lecithin 1% + Blood serum 0% (without serum), P2: Tris buffer + Soya Lecithin 1% + Blood serum 5%, P3: Tris buffer + Soya Lecithin 1% + Blood serum 10%. Treatment 3 with a blood serum concentration of 10% gave the best outcomes, with spermatozoa motility statistics showing a significant difference (P<0,05). The analysis of the plasma membrane integrity (MPU) parameters showed no significant difference (P>0,05).
