      Laju Pertumbuhan Biomassa Wickerhamomyces anomalus Menggunakan Media Limbah Cair Sari Kurma Sebagai Penghasil Se-yeast

      Abstract
      Selenium merupakan mikromineral esensial yang berfungsi sebagai antioksidan untuk melindung sel dari kerusakan radikal bebas. Pemberian selenium organik (Se-yeast) dinilai lebih aman karena memiliki bioavailabilitas tinggi, mudah diserap dan tingkat toksisitas yang rendah. fermentasi sel densitas tinggi (HCD) dirancang untuk menghasilkan biomassa dan produk dengan konsentrasi yang tinggi melalui pemberian sumber karbon konsentrasi tinggi. Penelitian ini bertujuan mengevaluasi laju pertumbuhan khamir Wickerhamomyces anomalus dengan waktu pemberian limbah cair sari kurma (LCSK) yang berbeda. Proses fermentasi dilakukan dengan metode HCD menggunakan media LCSK konsentrasi 70 gL-1 yang diperkaya sodium selenite 45 ppm dengan penambahan pada jam ke-10, 12, dan 24. Parameter yang diamati meliputi nilai pH, kadar glukosa, pertumbuhan biomassa dan kandungan selenium. Hasil penelitian menunjukkan bahwa waktu pemberian LCSK paling efektif terjadi pada jam ke-24, dengan nilai biomassa maksimal sebesar 1,58 gL-1 dan kadar selenium sebanyak 0,0364 mgL-1. Selain itu, pemberian LCSK pada saat sel starving dapat mempercepat laju pertumbuhan biomassa W. anomalus. Khamir W. anomalus mulai memasuki fase stasioner pada kisaran jam ke-42 dan pada akhir fermentasi sel belum mencapai fase kematian. Simpulan penelitian ini adalah pemberian LCSK konsentrasi tinggi pada jam ke-24 dapat meningkatkan laju pertumbuhan biomassa dan W. anomalus dapat mulai dipanen pada jam ke-42.
       
      Selenium is an essential micronutrient that functions as an antioxidant to protect cells from damage caused by free radicals. Organic selenium (Se-yeast) is considered safer due to their high bioavailability, better absorption, and lower toxicity. High Cell Density (HCD) fermentation is designed to produce biomass and products at high concentrations by supplying high-concentration carbon sources. This study aimed to evaluate the growth rate of Wickerhamomyces anomalus with different timing of extract date palm by-product (LCSK). Fermentation was conducted using the HCD method with 70 gL-1 LCSK medium enriched with 45 ppm sodium selenite. LCSK was added at different times i.e. the 10th, 12th, and 24th hour. The parameters measured included pH, glucose concentration, biomass growth, and selenium content. The results showed that the most effective LCSK addition occurred at hour 24, yielding the highest biomass at 1.58 gL-1 and selenium content of 0.0364 mgL-1. Supplementing LCSK during the cell starvation phase accelerated biomass accumulation. This yeast entered the stationary phase around 42 hours and by the end of fermentation, the cells had not yet reached the death phase. In conclusion, high-concentration LCSK addition at hour 24 enhances biomass growth rate and W. anomalus can be harvested starting at 42 hours.
