dc.description.abstractResistensi terhadap antibiotik konvensional menjadi tantangan besar dalam dunia kesehatan, sehingga diperlukan penemuan agen antimikroba baru. Antimicrobial peptide (AMP) merupakan kandidat alternatif yang menjanjikan karena memiliki risiko resistensi yang rendah. Salah satu AMP yang potensial adalah tripsin inhibitor yang dapat diperoleh dari tanaman Cucurbitaceae. Penelitian ini bertujuan mengisolasi dan mengidentifikasi gen penyandi AMP tripsin inhibitor dari lima spesies tanaman Cucurbitaceae, yaitu pare, melon, mentimun, labu kuning, dan labu siam. Isolasi DNA dilakukan dengan metode CTAB yang dimodifikasi. Identifikasi gen diawali dengan desain primer spesifik dari gen tripsin inhibitor-1 dan dilakukan amplifikasi gen dengan metode PCR. Hasil menunjukkan modifikasi isolasi dengan daun beku, penggunaan nitrogen cair, penambahan RNAse dan proteinase-K, serta CTAB 2% menghasilkan konsentrasi dan kemurnian DNA optimal yaitu 1,83?1,92 (A260/280) dan 2,02?2,52 (A260/230). Amplifikasi dilakukan pada suhu annealing 54,7°C dengan primer spesifik yang berhasil di desain. Amplifikasi menghasilkan pita DNA berukuran ~279 bp pada seluruh sampel. Hasil ini menunjukkan bahwa gen tripsin inhibitor-1 teridentifikasi pada kelima tanaman yaitu pare, melon, mentimun, labu kuning, dan labu siam. dc.description.abstractResistance to conventional antibiotics poses a major challenge in the medical field, highlighting the need for the discovery of new antimicrobial agents. Antimicrobial peptides (AMPs) are promising alternative candidates due to their low risk of resistance. One potential AMP is a trypsin inhibitor which can be derived from plants of the Cucurbitaceae. This study aimed to isolate and identify the gene encoding the trypsin inhibitor AMP from five Cucurbitaceae species: bitter melon, melon, cucumber, pumpkin, and chayote. DNA isolation was carried out using a modified CTAB method. Gene identification began with the design of specific primers for the trypsin inhibitor-1 gene, followed by gene amplification using the PCR method. The results showed that the modified isolation method using frozen leaves, liquid nitrogen, RNAse and proteinase-K, and 2% CTAB produced DNA with optimal concentration and purity, with A260/280 values ranging from 1.83 to 1.92 and A260/230 values from 2.02 to 2.52. Amplification was performed at an annealing temperature of 54.7°C using successfully designed specific primers. The amplification yielded ~279 bp DNA bands in all samples. These results indicate that the trypsin inhibitor-1 gene was identified in all five plant species: bitter melon, melon, cucumber, pumpkin, and chayote.
