dc.description.abstractIkan karang merupakan komoditas perikanan bernilai ekonomi tinggi di Indonesia, baik untuk konsumsi domestik maupun ekspor. Identifikasi ikan karang sering terkendala oleh kemiripan morfologi antarspesies, sehingga pendekatan genetik melalui DNA barcoding menjadi solusi untuk meningkatkan keakuratan identifikasi. Penelitian ini bertujuan mengidentifikasi ikan karang ekonomis tinggi menggunakan DNA barcoding berbasis gen Cytochrome Oxidase I (COI). Sebanyak 50 sampel ikan dianalisis melalui ekstraksi DNA, amplifikasi dengan primer universal FishF1-FishR1 menggunakan PCR, elektroforesis, serta sekuensing. Data sekuens dianalisis menggunakan BLAST pada GenBank (NCBI) dan BOLD system, sementara pohon filogeni dibangun dengan model Kimura 2-Parameter (K2P) menggunakan metode Neighbor-Joining (NJ) dengan 1000 bootstrap melalui MEGA 11. Hasil menunjukkan bahwa 49 sampel berhasil teridentifikasi mencakup 17 spesies, dengan genus Epinephelus dan Lutjanus mendominasi, serta membentuk tiga klad utama yang mencerminkan hubungan kekerabatan antarspesies. Identifikasi genetik ini memberikan kontribusi dalam pengelolaan sumber daya ikan karang secara berkelanjutan.
dc.description.abstractReef fish is a fishery commodity of high economic value in Indonesia, both for domestic consumption and export. The identification of reef fish is often constrained by morphological similarities between species, so the genetic approach through DNA barcoding is a solution to improve the accuracy of identification. This study aims to identify high-economy reef fish using DNA barcoding based on the Cytochrome Oxidase I (COI) gene. A total of 50 fish samples were analyzed through DNA extraction, amplification with FishF1-FishR1 universal primer using PCR, electrophoresis, and sequencing. Sequence data was analyzed using BLAST on GenBank (NCBI) and BOLD system, while phylogeny trees were constructed using the Kimura 2-Parameter (K2P) model and the Neighbor-Joining (NJ) method with 1000 bootstraps via MEGA 11. The results showed that 49 samples were successfully identified, covering 17 species dominated by Epinephelus and Lutjanus, and forming three main clades reflecting the kinship relationship between species. This genetic identification contributes to the sustainable management of coral fish resources.
