      Induksi Apoptosis oleh Ekstrak Air Daun Pepaya (Carica papaya L.) pada Sel Ragi (Saccharomyces cerevisiae)

      Abstract
      Daun pepaya (Carica papaya L.) diketahui memiliki aktivitas antioksidan yang dapat menurunkan kadar kolesterol pada penyakit jantung koroner. Oleh karena itu, diperlukan suatu strategi untuk mencegah pembentukan plak pada pembuluh darah melalui induksi apoptosis. Penelitian ini bertujuan mengidentifikasi kemampuan ekstrak air daun pepaya dalam menginduksi apoptosis pada sel ragi sebagai model sel eukariot. Ekstraksi dilakukan dengan metode dekokta, dan kandungan senyawa fitokimia dideteksi dengan uji kualitatif. Kemampuan ekstrak daun pepaya dalam menginduksi apoptosis pada Saccharomyces cerevisiae ditentukan dengan perhitungan frekuensi sel petite dan pengamatan aktivitas membran mitokondria melalui pencitraan fluoresensi. Analisis fitokimia menunjukkan bahwa ekstrak daun pepaya mengandung senyawa flavonoid, fenolik, tanin, alkaloid, dan saponin. Ekstrak air daun pepaya dengan konsentrasi 10 µg/mL memiliki frekuensi sel petite sebesar 73,92±17,38%, sedangkan pada konsentrasi 200 µg/mL sebesar 100,00±0,00%, dan memiliki aktivitas tertinggi dalam induksi apoptosis. Penentuan aktivitas mitokondria dilihat dari rendahnya intensitas pendaran warna jingga kemerahan yang dihasilkan, dimana sel telah mengalami disfungsi mitokondria dan terjadi apoptosis.
       
      Papaya leaves (Carica papaya L.) are known to have antioxidant activity that can lower cholesterol levels in coronary heart disease. Therefore, a strategy is needed to prevent plaque formation in blood vessels through apoptosis induction. The aim of the study was to identify the ability of papaya leaf water extract to induce apoptosis in yeast cells as a model of eukaryotic cells. Extraction was carried out using the decoction method, and the phytochemical compound content was detected by qualitative testing. The ability of papaya leaf extract to induce apoptosis in Saccharomyces cerevisiae was determined by calculating the frequency of petite cells and observing mitochondrial membrane activity through fluorescent imaging. Phytochemical analysis showed that papaya leaf extract contains flavonoids, phenolics, tannins, alkaloids, and saponins. Papaya leaf water extract with a concentration of 10 µg/mL had a petite cell frequency of 73.92±17.38%, while at a concentration of 200 µg/mL it was 100.00±0.00%, and had the highest activity in apoptosis induction. Determination of mitochondrial activity is seen from the low intensity of the reddish orange fluorescence produced, where the cells have experienced mitochondrial dysfunction and apoptosis has occurred.
