      Identifikasi Keanekaragaman Fitoplankton Menggunakan Metode Konvensional dan DNA Lingkungan di Perairan Selat Malaka

      Abstract
      Selat Malaka merupakan perairan strategis yang dipengaruhi oleh dinamika arus permukaan, yang berdampak pada distribusi nutrien. Nutrien ini berperan penting dalam kelimpahan dan keanekaragaman fitoplankton, yang merupakan dasar rantai makanan di ekosistem laut serta indikator kesehatan perairan. Penelitian ini bertujuan mengidentifikasi keanekaragaman dan struktur komunitas fitoplankton di perairan Selat Malaka. Fitoplankton di identifikasi menggunakan dua metode yaitu metode konvensional (analisis mikroskopis) dan metode e-DNA (analisis genetik berbasis ekstraksi DNA Lingkungan). Identifikasi menggunakan metode konvensional menemukan 43 spesies fitoplankton dengan nilai indeks keanekaragaman dalam kategori sedang di seluruh titik stasiun. Indeks keseragaman tergolong sedang hingga tinggi, dengan Trichodesmium sp. sebagai spesies yang paling dominan. Sebaliknya, metode e-DNA berhasil mendeteksi 262 spesies fitoplankton, dengan nilai indeks keanekaragaman dan keseragaman yang termasuk kategori tinggi di seluruh titik stasiun, tanpa spesies yang mendominasi.
       
      The Strait of Malacca is a strategic water influenced by the dynamics of surface currents, which have an impact on the distribution of nutrients. These nutrients play an important role in the abundance and diversity of phytoplankton, which are the basis of the food chain in marine ecosystems and indicators of aquatic health. This study aims to identify the diversity and structure of phytoplankton communities in the waters of the Strait of Malacca. Phytoplankton were identified using two methods, namely the conventional method (microscopic analysis) and the e-DNA method (genetic analysis based on Environmental DNA extraction). Identification using the conventional method found 43 phytoplankton species with diverse index values in the moderate category at all station points. The evenness index is classified as moderate to high, with Trichodesmium sp. as the most dominant species. In contrast, the e-DNA method successfully detected 262 phytoplankton species, with diversity and evenness index values in the high category at all station points, without a dominant species.
