      Isolasi, Identifikasi, dan Karakterisasi Cendawan Dark Septate Endophyte (DSE) pada Jabon Merah (Neolamarckia macrophylla (Wall.) Bosser.

      Abstract
      Penurunan produktivitas hutan di Indonesia mendorong perlunya pendekatan inovatif dalam rehabilitasi ekosistem hutan. Upaya tersebut menghadapi berbagai tantangan, termasuk ketersediaan bibit berkualitas. Cendawan endofit bersepta gelap (dark septate endophyte/DSE) menjadi salah satu solusi yang menjanjikan dalam meningkatkan kualitas dan ketahanan tanaman. Akar pohon jabon merah (Neolamarckia macrophylla) dinilai memiliki asosiasi yang potensial dengan cendawan DSE. Penelitian ini bertujuan mengidentifikasi dan mengkarakterisasi cendawan DSE pada akar pohon jabon merah, serta mengkaji sifat isolat DSE terhadap pertumbuhan tanaman. Sampel akar dikumpulkan dari 25 pohon terpilih di Parungpanjang, Bogor kemudian dianalisis untuk kolonisasi dan isolasi, diikuti dengan karakterisasi makroskopis, mikroskopis, dan identifikasi molekuler pada isolat DSE. Tujuh isolat DSE (MPS-1, MPM-2, MPM-3, MPS-4, MPM-5, MPS-6, MPM-7) berhasil diisolasi dan diidentifikasi dengan keragaman sifat makroskopis dan mikroskopisnya. Hasil identifikasi molekuler mengungkapkan hubungan erat isolat tersebut dengan spesies dalam filum Ascomycota, seperti Neophaeocryptopus spartii, Parawiesneriomyces chiayiensis, dan Pleosporales sp. Benih Brassica pekinensis digunakan dalam uji pertumbuhan tanaman secara in vitro, kemudian diinokulasi dengan ketujuh isolat DSE. Hasil pengamatan menunjukkan peningkatan panjang total bibit secara signifikan pada enam perlakuan isolat DSE dibandingkan dengan kontrol. Penelitian lebih lanjut diperlukan untuk mengeksplorasi kandungan metabolit sekunder, efektivitas jangka panjang, interaksi antarspesies, dan aplikasinya di lapangan guna mengoptimalkan penggunaan DSE sebagai pupuk hayati.
       
      The decline in the productivity of forests in Indonesia underscores the need for innovative approaches to forest rehabilitation. One critical challenge in such efforts is the availability of high-quality seedlings. Dark septate endophyte (DSE) fungi have emerged as a promising solution for enhancing plant quality and resilience. The roots of red jabon trees (Neolamarckia macrophylla) are considered to have potential associations with DSE fungi. This study aimed to identify and characterize DSE fungi associated with the roots of red jabon trees and to evaluate the properties of DSE isolates in promoting plant growth. Root samples were collected from 25 selected trees in Parungpanjang, Bogor, analyzed for colonization and isolation, followed by macroscopic and microscopic characterization, as well as molecular identification of the DSE isolates. Seven DSE isolates (MPS-1, MPM-2, MPM-3, MPS-4, MPM-5, MPS-6, MPM-7) were successfully isolated and identified, exhibiting diverse macroscopic and microscopic characteristics. Molecular identification revealed a close relationship between the isolates and species within the phylum Ascomycota, such as Neophaeocryptopus spartii, Parawiesneriomyces chiayiensis, and Pleosporales sp. Seeds of Brassica pekinensis were used in an in vitro plant growth assay. Seedlings inoculated with six of the seven DSE isolates showed a significant increase in total seedling length compared to the control. Further research is needed to explore secondary metabolite content, long-term effectiveness, interspecies interactions, and field applications to optimize the use of DSE as biofertilizers.
