dc.description.abstract | Spodoptera litura merupakan hama polifag penting pada beberapa jenis tanaman budidaya sehingga keberadaannya perlu dikendalikan. Salah satu metode pengendalian ramah lingkungan adalah menggunakan entomopatogen, Spodoptera litura nucleopolyhedrovirus (SpltNPV). Virus ini bersifat spesifik inang dan dinilai cukup efektif mengendalikan S. litura. Genom SpltNPV mengandung gen yang bertanggung jawab dalam infeksi oral yaitu Per Os Infectivity Factors-3 (pif-3). Gen tersebut ditemukan pada sebagian besar Baculovirus sehingga karakterisasi molekuler dapat dilakukan. Oleh karena itu, penelitian ini bertujuan mengidentifikasi karakter molekuler berdasarkan gen pif-3 untuk mengetahui kekerabatan SpltNPV isolat Bogor dengan isolat NPV lainnya. Amplifikasi fragmen DNA SpltNPV dilakukan dengan metode Polymerase Chain Reaction (PCR). Ukuran fragmen DNA SpltNPV yang berhasil diamplifikasi yaitu sebesar 720 pb. Pengurutan gen pif-3 SpltNPV isolat Bogor berdasarkan nukleotida diperoleh ukuran sebesar 603 pb dan 200 asam amino. Berdasarkan hasil analisis homologi dan filogenetik, SpltNPV isolat Bogor berkerabat dekat dengan SpltNPV dan SpltMNPV asal Jepang dan Cina. Nilai homologi terbesar yang diperoleh berdasarkan urutan nukleotida hasil amplifikasi adalah 99,80%, sedangkan berdasarkan urutan asam amino pengkode pif-3 adalah sebesar 100%. Konstruksi pohon filogenetik menunjukkan bahwa SpltNPV isolat Bogor termasuk dalam kelompok NPV yang menginfeksi spesies Spodoptera litura. | |
dc.description.abstract | Spodoptera litura is an important polyphagous pest of several cultivated plants that needs to be controlled. One environmentally friendly control method is using an entomopathogen, Spodoptera litura nucleopolyhedrovirus (SpltNPV). This virus is host-specific and considered quite effective in controlling S. litura. The SpltNPV genome contains one of the genes responsible for oral infection, called Per Os Infectivity Factors-3 (pif-3). This gene is found in most Baculoviruses, allowing for molecular characterization. Therefore, this study aims to identify molecular characters based on the pif-3 gene to determine the genetic similarity of SpltNPV Bogor isolate with other NPV isolates. Amplification of SpltNPV DNA fragments was carried out by Polymerase Chain Reaction (PCR) method. The size of the amplified SpltNPV DNA fragment was 720 bp. Sequencing of the pif-3 gene from SpltNPV Bogor isolate based on nucleotides obtained a size of 603 bp and 200 amino acids. Based on the results of homology and phylogenetic analysis, SpltNPV Bogor isolate is closely related to SpltNPV and SpltMNPV from Japan and China. The largest homology value obtained based on the amplified nucleotide sequence was 99,80%, while based on the amino acid sequence of pif-3 was 100%. The phylogenetic tree constructed showed that SpltNPV Bogor isolate belongs to a group of NPVs infecting the Spodoptera litura species. | |
