      Abstract
      RINGKASAN DIAN CIPTA RINI. Aktivitas Antitumor Nanoemulsi Ekstrak Etanol Typhonium flagelliforme pada Tikus yang Diinduksi DMBA. Dibimbing oleh BAMBANG PONTJO PRIOSOERYANTO, RIKI SISWANDI, dan LINA NOVIYANTI SUTARDI. Kanker diyakini sebagai penyakit penyebab kematian yang paling umum pada anjing peliharaan dan anjing kerja di negara berkembang. Penelitian ini bertujuan untuk mempelajari aktivitas antitumor dari nanoemulsi Typhonium flagelliforme pada tikus Sprague Dawley yang diinduksi dengan karsinogen 7,12- dimethylbenz(a)anthracene (DMBA). Nanoemulsi ekstrak etanol T. flagelliforme memiliki ukuran partikel 145,7 nm dan indeks polidispersitas 0,368. Ekstrak ini juga mengandung senyawa fitokimia seperti alkaloid, flavonoid, steroid, terpenoid, tanin, dan saponin. Uji toksisitas menggunakan metode brine shrimp lethality test (BSLT) menunjukkan bahwa LC50 dari nanoemulsi T. flagelliforme adalah 384,41 ppm. Sebanyak 25 ekor tikus dibagi menjadi lima kelompok, dan setiap tikus diinjeksi subkutan dengan DMBA (25 µg/0,05 mL) dua kali seminggu selama 12 minggu. Kelompok K1 tidak menerima perlakuan setelah induksi DMBA; kelompok K2 diinjeksi dengan plasebo (0,05 mL pelarut nanoemulsi); kelompok K3 diinjeksi 25 µg/kg BB nanoemulsi T. flagelliforme; kelompok K4 diinjeksi 50 µg/kg BB nanoemulsi T. flagelliforme; dan kelompok K5 diinjeksi 100 µg/kg BB nanoemulsi T. flagelliforme. Terbentuk empat jenis tumor kulit yang dikonfirmasi melalui observasi makroskopik dan histopatologis yaitu anaplastic carcinoma, myosarcoma, squamous cell carcinoma (SCC), dan fibrosarcoma. Perlakuan diberikan secara intratumoral, tiga hari dalam seminggu selama empat minggu. Hasil menunjukkan bahwa ukuran tumor pada kelompok yang diberi nanoemulsi T. flagelliforme (K3, K4, K5) mengalami penurunan signifikan (p < 0,05) dibandingkan dengan kelompok K1. Indeks mitosis dan angiogenesis juga menurun pada kelompok yang diberi nanoemulsi. Tidak ada perubahan signifikan dalam tes hematologi dan kimia darah, yang menunjukkan bahwa terapi nanoemulsi tidak mempengaruhi sistem hematopoietik. Penelitian ini menyimpulkan bahwa ekstrak etanol daun T. flagelliforme dalam bentuk nanoemulsi adalah kandidat menjanjikan untuk pengobatan antitumor pada tumor kulit dan dosis terbaik dalam studi ini adalah 100 µg/kg BB.       SUMMARY DIAN CIPTA RINI. Antitumor Activity of Typhonium flagelliforme Ethanol Extract in Nanoemulsion Form in DMBA-Induced Rats. Cancer is believed to be the most common cause of disease-related death in companion and working dogs in developing countries. This research aimed to study the antitumor activity of Typhonium flagelliforme nanoemulsion in Sprague Dawley rats induced by the carcinogen 7,12-dimethylbenz(a)anthracene (DMBA). The nanoemulsion of T. flagelliforme ethanol extract had a particle size of 145,7 nm and a polydispersity index of 0,368. It also contained phytochemical compounds such as alkaloids, flavonoids, steroids, terpenoids, tannins, and saponins. A toxicity test using the brine shrimp lethality method showed that the LC50 of T. flagelliforme nanoemulsion is 384.41 ppm. A total of 25 rats were divided into five groups, and each rat was injected subcutaneously with DMBA (25 µg/0.05 mL) twice weekly for 12 weeks. The K1 group did not receive any treatment after DMBA induction; the K2 group was injected with a placebo (0.05 mL nanoemulsion solvent); the K3 group was treated with 25 µg/kg BW of T. flagelliforme nanoemulsion; the K4 group was treated with 50 µg/kg BW of T. flagelliforme nanoemulsion; and the K5 group was treated with 100 µg/kg BW of T. flagelliforme nanoemulsion. Four types of tumors were formed in the skin, confirmed through macroscopic and histopathological observation: anaplastic carcinoma, myosarcoma, squamous cell carcinoma, and fibrosarcoma. The treatments were administered intratumorally, three days a week for four weeks. Results showed that the tumor size in the treated groups (K3, K4, K5) significantly decreased after four weeks of T. flagelliforme nanoemulsion treatment (p < 0.05) compared to the K1 group. The mitosis and angiogenesis indices also decreased in the treatment groups. There were no significant changes in the hematological and blood chemistry tests, which showed that the nanoemulsion therapy did not affect the hematopoietic system. This study concludes that the ethanol extract of T. flagelliforme leaves in nanoemulsion form is a promising candidate for antitumor treatment in skin tumors and the best dose in this study is 100 µg/kg BW.
