      Gambaran Hematologi Pada Tikus Model Strok Dengan Metode Middle Cerebral Artery Occlusion

      Abstract
      Strok merupakan gejala kelumpuhan saraf akibat gangguan aliran darah pada bagian otak. Strok iskemik termasuk jenis strok yang sering terjadi pada masyarakat. Penelitian ini bertujuan untuk menganalisis hematologi pada hewan coba yang diinduksi strok iskemik dengan metode middle cerebral artery occlusion (MCAO). Penelitian ini menggunakan 8 ekor tikus galur Sprague-dawley jantan, dibagi menjadi 2 kelompok, yaitu kontrol dan perlakuan. Tikus yang di induksi strok diamati menggunakan penilaian tes Bederson. Darah diambil dari jantung untuk dianalisis terhadap parameter hematologi. Hasil penelitian menunjukkan penilaian dengan skor Bederson 1 yang ditandai fleksi kontralateral cedera otak kiri. Jumlah sel darah putih (WBC) pada kelompok kontrol dan perlakuan berturut-turut adalah 5,3 x 103/mm3 dan 9,1 x 103/mm3. Jumlah sel darah merah (RBC) pada kelompok kontrol dan perlakuan adalah 7,7 x 106/mm3 dan 4,8 x 106/mm3. Konsentrasi hemoglobin pada kelompok kontrol dan perlakuan adalah 14,2 g/dL dan 8,7 g/dL. Nilai hematokrit pada kelompok kontrol dan perlakuan adalah 42,8 % dan 26,3 %. Sebagai simpulan, penerapan metode MCAO pada tikus penelitian menyebabkan kerusakan neurologis ringan, meningkatkan jumlah sel darah putih, serta menurunkan jumlah sel darah merah, konsentrasi hemoglobin, dan nilai hematokrit.
       
      Stroke is a symptom of nerve paralysis due to impaired blood flow in the brain. Ischemic stroke is a type of stroke whose symptoms often occur in the community. This study aims to analyze hematology by inducing ischemic stroke in experimental animals with the middle cerebral artery occlusion (MCAO) method to determine whether ischemic stroke affects hematological values. This study used 8 male Sprague-Dawley rats, divided into 2 groups, namely, control and treatment. Rats induced by stroke were observed using the Bederson test assessment. Blood was taken from the heart to be analyzed for hematological parameters. The results showed an assessment with a Bederson score of 1 characterized by contralateral flexion of the left brain damage. The number of white blood cells (WBC) in the control and treatment groups were 5,3 x 103/mm3 and 9,1 x 103/mm3, respectively. The number of red blood cells (RBC) in the control and treatment groups were 7,7 x 106/mm3, 4,8 x 106/mm3. Hemoglobin levels in the control and treatment groups were 14.2 g/dL, 8.7 g/dL. Hematocrit values in the control and treatment groups were 42.8%, 26.3%. In conclusion, the application of the MCAO method to the research rats caused mild neurological damage, increased the number of white blood cells, and decreased the number of red blood cells, hemoglobin levels, and hematocrit values.
