      Gambaran Histopatologi Pankreas Tikus Jantan yang Diberi Karsinogen DMBA dan Diobati Ekstrak Nanopartikel Thyphonium flagelliforme

      Abstract
      Tumor merupakan suatu gangguan pertumbuhan yang ditandai dengan adanya proliferasi atau perbanyakan sel yang berlebih, abnormal, dan tidak terkontrol. Pengobatan tumor sudah banyak dikembangkan, salah satunya adalah pengobatan herbal. Penelitian ini bertujuan menganalisa efek pengobatan tumor menggunakan ekstrak nanopartikel Typhonium flagelliforme pada gambaran histopatologi pankreas. Penelitian ini menggunakan 26 ekor tikus jantan yang diinjeksi karsinogen DMBA (7,12-dimethylben[a]anthracene) secara subkutan dengan dosis 25 µg/0,05 ml/ekor dua kali seminggu selama empat minggu. Tikus dibagi menjadi 5 kelompok perlakuan yaitu: kontrol negatif; pelarut nanopartikel berupa etanol 0,1 ml; dan tiga kelompok perlakuan injeksi ekstrak nanopartikel keladi tikus dengan dosis 25 µg; 50 µg; dan 100 µg secara intratumoral. Tikus diberi perlakuan injeksi nanopartikel ekstrak keladi tikus tiga kali seminggu selama empat minggu. Tikus dieutanasia kemudian pankreas dikoleksi dan dibuat preparat histopatologi dengan pewarnaan Hematoksilin–Eosin (HE) dan diamati menggunakan mikroskop dengan pembesaran 20x. Variabel yang diamati adalah jumlah sel nekrosis pada pankreas. Hasil uji Kruskall-Wallis menunjukkan tidak ada perbedaan nyata (p > 0,05) pada jumlah sel nekrosa pankreas di kelima kelompok perlakuan. Hasil penelitian ini menunjukkan bahwa perubahan patologis pankreas tikus jantan akibat pemberian DMBA secara subkutan di area punggung berupa karyopiknosis dengan jumlah yang terbatas. Pemberian ekstrak nanopartikel Typhonium flagelliforme tidak berpengaruh nyata pada gambaran histopatologi pankreas tikus yang diinduksi karsinogen DMBA secara subkutan.
       
      Tumor is a growth disorder characterized by the excessive, abnormal, and uncontrolled proliferation or multiplication of cells. Treatments for tumors have been widely developed, one of which is herbal treatment. This study aimed to analyze the effect of treating tumors using Typhonium flagelliforme nanoparticle extract on pancreatic histopathological features. The study involved 26 male rats injected subcutaneously with the carcinogen DMBA (7,12- dimethylbenz[a]anthracene) at a dose of 25 µg/0.05 ml/rat twice a week for four weeks. The rats were divided into 5 treatment groups: negative control; ethanol 0.1 ml; and three treatment groups with Typhonium flagelliforme nanoparticle extract injections at doses of 25 µg, 50 µg, 100 µg intratumorally. The rats were treated with nanoparticle extract injections three times a week for four weeks. After euthanasia, the pancreases were collected and histopathological preparations were made using Hematoxylin-Eosin (HE) staining, observed under a microscope at 20x magnification. The variable observed was the number of necrotic cells in pancreas. The Kruskal-Wallis test results showed no significant difference (p > 0.05) in the number of necrotic cells in pancreas among the five treatment groups. The results of this study indicate that histopathological changes in the pancreas of male rats that have been induced by DMBA via subcutaneous administration in the dorsal area include karyopyknosis. The administration of Typhonium flagelliforme nanoparticle extract had no significant effect on the histopathology of the pancreas in rats induced with the DMBA carcinogen
