      Karakteristik Gen 16S rRNA Anura (Famili: Dicroglossidae) di Sekitar Danau SDGs IPB Sebagai Dasar Konservasi

      Abstract
      Anura atau katak dan kodok merupakan ordo yang sensitif terhadap perubahan lingkungan perairan karena fase larvanya yang bergantung pada kualitas perairan sehingga Anura dapat dijadikan sebagai bioindikator yang baik. Salah satu genus dari famili Dicroglossidae yang digunakan yaitu Fejervarya merupakan spesies kriptik sehingga diperlukan identifikasi molekuler. Penelitian ini bertujuan mengidentifikasi karakteristik gen famili Dicroglossidae di sekitar Danau SDGs IPB berdasarkan marka gen 16S rRNA sebagai dasar informasi untuk konservasi genetik. Penelitian dilaksanakan pada Juli 2023 di Danau SDGs IPB menggunakan metode Visual Encounter Survey. Analisis data yang dilakukan yaitu DNA barcoding, validasi spesies, analisis jarak genetik, filogeni, dan diversitas haplotipe. Contoh berhasil diidentifikasi sebagai Fejervarya cancrivora, Fejervarya limnocharis, dan Limnonectes macrodon. Hubungan kekerabatan antarcontoh erat, keragaman genetik interspesies tinggi namun intraspesies rendah sehingga diperlukan pengelolaan habitat Dicroglossidae lebih lanjut di Danau SDGs IPB.
       
      Anura (frogs and toads), an order highly sensitive to aquatic environment changes due to its reliance on water quality during the larval phase, serves as an effective bioindicator. Within the Dicroglossidae family, the genus Fejervarya is cryptic so molecular identification is required. This study aims to characterize the genes of the Dicroglossidae family around SDGs Lake in IPB using 16S rRNA gene markers to provide foundational genetic conservation information. The research was conducted in July 2023 at SDGs Lake IPB using Visual Encounter Survey method. Data analyses included DNA barcoding technique, species validation, genetic distance analysis, phylogeny, and haplotype diversity. Successful identification of samples as Fejervarya cancrivora, Fejervarya limnocharis, and Limnonectes macrodon reveals close relationships and high interspecies genetic diversity but low intraspecies diversity, indicating the need for further habitat management of the Dicroglossidae family at SDGs Lake, IPB.
