      Respons Imun Udang Vaname yang Dipelihara dengan Kepadatan Berbeda pada Sistem Bioflok terhadap Infeksi Vibrio parahaemolyticus

      Abstract
      Budidaya sistem intensif pada udang vaname dengan kepadatan tinggi dapat menyebabkan udang stress dan mudah terinfeksi penyakit bakterial, yang salah satunya adalah Vibrio parahaemolyticus. Pengendalian infeksi bakteri V. parahaemolyticus dapat dilakukan dengan penentuan padat tebar yang optimal menggunakan sistem budidaya berbasis bioflok. Sistem bioflok diterapkan melalui penambahan sumber karbon organik untuk mengonversi nitrogen menjadi biomassa mikroorganisme heterotrof karena mengandung senyawa ekstraseluler yang dapat memodulasi respons imun dan menurunkan virulensi bakteri. Penelitian ini bertujuan untuk menganalisis respons imun dan resistansi udang vaname pada tahap awal pendederan yang dipelihara dengan padat tebar berbeda pada sistem budidaya berbasis bioflok terhadap infeksi V. parahaemolyticus. Udang vaname yang digunakan memiliki bobot awal rata-rata 0,44±0,02 g/ekor. Udang dipelihara selama 7 hari dengan masing-masing perlakuan meliputi sistem bioflok dan tanpa sistem bioflok sebagai kontrol dibandingkan pada empat padat tebar yaitu 4, 8, 12, 16 ekor L-1 yang diuji tantang secara perendaman dalam media menggunakan V. parahaemolyticus dengan dosis 105 CFU mL-1. Sistem bioflok mampu meningkatkan respons imun, menurunkan kelimpahan populasi sel V. parahaemolyticus di air pemeliharaan dan tubuh udang, serta meningkatkan tingkat kelangsungan hidup udang vaname hingga padat tebar tertinggi (16 ekor L-1).
       
      Intensive system cultivation of white shrimp with high density can cause stress and shrimp easily affected with bacterial infection, one of which is Vibrio parahaemolyticus. Control of V. parahaemolyticus bacterial infection can be done by determining the optimal stocking density using a biofloc-based cultivation system. The biofloc system is applied through the addition of organic carbon sources to convert nitrogen into heterotrophic microorganisms biomass because it contains extracellular compounds that can modulate immune responses and reduce bacterial virulence. This study aims to analyze the shrimp’s immune response and resistance in the early stages of the biofloc-based rearing system to V. parahaemolyticus infection. The test shrimp used had an initial average weight of 0,44±0,02 g each. The shrimp were reared for 7 days with different stocking densities. Shrimp were reared for 7 days with each treatment including a biofloc system and without a biofloc system as a control compared to four stocking densities of 4, 8, 12, 16 fish L-1 which were challenged using V. parahaemolyticus via immersion at a dose of 105 CFU mL-1. The biofloc system was able to increase the immune response, reduce the abundance of V. parahaemolyticus cells population in rearing water and shrimp bodies, as well as increase the survival rate of white shrimp to the highest stocking density (16 shrimp L-1).
