      Potensi Ekstrak Kayu Bajakah (Uncaria spp.) dalam Regulasi Aktivitas GLUT-4 pada Penderita Resistensi Insulin

      Abstract
      Kayu bajakah (Uncaria sp.) diyakini oleh beberapa kalangan masyarakat sebagai obat bagi berbagai penyakit, tak terkecuali diabetes melitus tipe 2. Akan tetapi, penelitian akan kayu bajakah sebagai antidiabetes masih belum banyak dilakukan. Identifikasi ekstrak kayu bajakah melalui LC-MS/MS diharapkan mendapat senyawa potensial dalam ekstrak kayu bajakah sebagai penanganan resistensi insulin dan diabetes melitus tipe 2. Penelitian ini bertujuan mengidentifikasi senyawa aktif kayu bajakah dan potensinya dalam persinyalan insulin untuk meningkatkan kerja GLUT-4 melalui penambatan protein PTEN. Ekstraksi dilakukan dengan menggunakan metode maserasi dan dianalisis menggunakan LC-MS/MS dan diprediksi secara in silico. Terdeteksi sebanyak 14 senyawa aktif pada ekstrak kayu bajakah dan prediksi melalui penambatan molekuler, toksisitas, dan bioavailabilitas menunjukkan satu senyawa potensial, yaitu pellitorin. Simpulan penelitian ini adalah pellitorin mampu membentuk interaksi yang kuat pada tiga loop (P loop, WDP loop, dan TI loop) yang menyusun sisi aktif PTEN. Terinhibisinya PTEN akan meningkatkan efektivitas metabolisme persinyalan insulin dan meningkatkan kinerja GLUT4.
       
      Traditionally, bajakah wood (Uncaria sp.) has been used to treat various ailments, including type 2 diabetes mellitus. However, scientific research on its antidiabetic properties is scarce. This study aimed to identify active compounds within bajakah wood extract and their potential to improve insulin signaling and GLUT-4 function in managing insulin resistance and type 2 diabetes mellitus. Bajakah wood was extracted using maceration method. The extract was the analyzed using LC-MS/MS and in silico prediction was employed to assess the potential of these identified compounds for molecular docking, toxicity, and bioavailability. The analysis revealed a total of 14 active compounds within bajakah wood extract. Through in silico prediction, pellitorin emerged as a promising compund. The conclution of this study was that pellitorin demonstrated a strong binding affinity to three specific loops that form the active site of PTEN (P loop, WDP loop, and TI loop). The inhibition of PTEN enhances the effectiveness of insulin signaling metabolism and improves GLUT-4 function.
