      Aplikasi Ekstrak Ketapang Terminalia catappa sebagai Antimikroba pada Pendederan Botia India Botia lohachata

      Abstract
      Daun ketapang dapat memberikan efek antibakteri dan antijamur. Penelitian terapan ini bertujuan untuk mengetahui aplikasi ekstrak daun ketapang sebagai antimikroba pada budidaya ikan botia. Penelitian ini terdiri dari uji in vitro dan uji in vivo. Pada uji in vitro dilakukan pengujian diameter zona hambat ketapang terhadap bakteri Aeromonassp. dengan perlakuan kontrol, rebus 30 menit, alat Pressured Hot Water Extraction (PHWE) 30 menit, dan PHWE 45 menit dengan 5 pengulangan. Hasil menunjukkan beda nyata (P<0,5) pada daun ketapang yang diekstraksi alat PHWE 45 menit dengan kadar total fenol pada sampel ekstrak ketapang sebanyak 5,1 mg GAE/g ketapang. Aplikasi metode terbaik pada ikan botia dengan penggunaan dosis ekstrak ketapang 0,04 mL/L, dosis 0,8 mL/L, dan kontrol. Nilai intensitas dan prevalensi ektoparasit pada perlakuan aplikasi ekstrak ketapang 0,8 mL/L dengan rata-rata nilai 0,56±1,13 ind/ekor dan 22,22±19,25%. Kelimpahan total bakteri pada perlakuan aplikasi ekstrak ketapang 0,8 mL/L sebesar 1,58±0,62×103 CFU/mL sedangkan kelimpahan bakteri Aeromonas sp memiliki nilai 9,75±0,54×102 CFU/mL. Perlakuan aplikasi ekstrak ketapang 0,8 mL/L memiliki sintasan sebesar 98,79±0,01% dan pertambahan panjang total 1,01±0,14 cm. Pertambahan panjang total pada perlakuan ekstrak ketapang 0,04 mL/L sebesar 0,78±0,11 cm. Kualitas air pemeliharaan dalam rentang normal. Aplikasi ekstrak ketapang 0,8 mL/L memiliki kecenderungan hasil yang lebih baik sebagai antimikroba pada pendederan ikan botia. Kata kunci: antimikroba, bakteri, botia, ketapang, parasit
       
      Ketapang leaves can provide antibacterial and antifungal effects. This study aimed to determine the application of ketapang leaf extract as an antimicrobial in botia fish farming. This research consists of in vitro tests and in vivo tests. In the in vitro test, the diameter of the inhibition zone of ketapang against Aeromonas sp. bacteria was tested with a control treatment, boiled for 30 minutes, 30 minutes Pressured Hot Water Extraction (PHWE) tool, and 45 minutes PHWE with five repetitions. The results showed a significant difference (P<0.5) in ketapang leaves extracted by PHWE 45 minutes with total phenol content in the ketapang extract sample of 5.1 mg GAE/g ketapang. Application of the best method on botia fish with the use of ketapang extract dose of 0.04 mL/L, dose of 0.8 mL/L, and control. The intensity and prevalence of ectoparasites in the treatment of 0.8 mL/L ketapang extract application with an average value of 0.56±1.13 ind/head and 22.22±19.25%. The total abundance of bacteria in the treatment of 0.8 mL/L ketapang extract application was 1.58±0.62×103 CFU/mL while the abundance of Aeromonas sp. bacteria had a value of 9.75±0.54×102 CFU/mL. The application treatment of ketapang extract 0.8 mL/L had a survival rate of 98.79±0.01% and a total length gain of 1.01±0.14 cm. The total length gain in the treatment of 0.04 mL/L ketapang extract was 0.78±0.11 cm. The water quality was maintained within the normal range. The application of 0.8 mL/L ketapang extract has a tendency to produce better results as an antimicrobial in botia fish breeding.
