      Efektivitas Ekstrak Buah Jambu Biji (Psidium guajava L.) terhadap Viabilitas Sel Kanker Kulit (Melanoma) dengan Microtetrazolium Assay

      Abstract
      Buah jambu biji (Psidium guajava L.) merupakan bahan alam yang memiliki senyawa aktif dengan potensi sebagai alternatif antikanker yang dipercaya memiliki efek samping minimal. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak buah jambu biji terhadap viabilitas sel kanker kulit melanoma secara in vitro melalui nilai persentase viabilitas, nilai persentase inhibisi dan perubahan morfologi sel. Metode penelitian yang dilakukan yaitu eksperimental. Tahap penelitian meliputi analisis perubahan morfologi sel, pengujian MTT assay untuk melihat pengaruh ektrak buah jambu biji (Psidium guajava L.) dengan beberapa tingkatan konsentrasi (1600; 800; 400; 200; 100; 50; 25; dan 12,5 µg/ml) terhadap viabilitas sel melanoma B16-F10 (ATCC CRL-6475) dan perhitungan nilai IC50 (inhibition concentration) menggunakan analisis regresi linear. Hasil penelitian menunjukkan kemampuan ekstrak buah jambu biji dalam memengaruhi viabilitas sel melanoma ialah 27,62% berdasarkan nilai inhibisi setelah pemberian ekstrak pada konsentrasi 1600 µg/ml dengan nilai IC50 sebesar 3801,30 µg/ml. Kemampuan sitotoksisitas juga ditunjukkan dengan apoptosis pada sel.
       
      Guava fruit (Psidium guajava L.) is a natural ingredient that has active compounds with the potential to be an anti-cancer alternative which is believed to have minimal side effects. This study aims to determine the effectiveness of guava fruit extract on the viability of melanoma skin cancer cells in vitro through viability percentage values, inhibition percentage values and changes in cell morphology. The research method used was experimental. The research stage includes analysis of changes in cell morphology, MTT assay testing to see the effect of guava fruit extract with several concentration levels (1600; 800; 400; 200; 100; 50; 25; and 12,5
