dc.description.abstract | Industri tekstil merupakan salah satu sektor industri yang paling penting dan
menempati urutan pertama ekspor non migas di Indonesia. Industri ini mengalami
perkembangan yang sangat pesat dan memberikan kontribusi yang besar terhadap
perekonomian dan kesejahteraan masyarakat. Namun, industri tekstil menghadapi
tantangan besar terutama dalam pengelolaan limbahnya. Limbah tekstil (limbah
cair tekstil) merupakan limbah yang sangat kompleks mengandung berbagai
senyawa kimia berbahaya, seperti zat warna sintetis, logam berat, dan bahan
organik lainnya yang berpotensi mencemari lingkungan jika dilepaskan tanpa
diolah terlebih dahulu. Dua zat warna sintetis yang paling banyak digunakan
dalam industri tekstil adalah grup antraquinon dan azo. Zat warna tersebut bersifat
racun, karsinogenik, dan berbahaya untuk lingkungan dan kesehatan manusia.
Oleh karena itu, diperlukan teknologi pengolahan limbah tekstil sebagai upaya
pengurangan pencemaran lingkungan. Pengolahan limbah tekstil berbasis
cendawan (mikoremediasi) atau tanaman (fitoremediasi) telah banyak dilaporkan.
Berbagai jenis cendawan dan tanaman pun telah banyak digunakan sebagai agen
bioremediasi limbah tekstil. Namun, sampai saat ini belum ada satu pun penelitian
terkait kemampuan cendawan Dark Septate Endophyte (DSE) dan tanaman
mangrove dalam meremediasi limbah tekstil termasuk dekolorisasi zat warna dan
penyisihan logam berat kromium VI (Cr(VI)).
Penelitian ini bertujuan: 1) Mengkarakterisasi DSE dengan potensi sebagai
pendegradasi zat warna sintetis (azo dan antraquinon) dan penyisih logam berat
Cr(VI). 2) Menganalisis faktor-faktor lingkungan yang mempengaruhi aktivitas
DSE dalam mendekolorisasi zat warna sintetis golongan azo dan antraquinon. 3)
Menemukan metode mikoremediasi yang sesuai melalui sistem sel terimobilisasi
dan konsorsium DSE untuk mendegradasi limbah tekstil. 4) Menguji coba
kombinasi sistem integrasi antara cendawan DSE dengan Floating Treatment
Wetland (FTW) menggunakan tanaman mangrove (Rhizophora sp.) dalam
mendegradasi limbah tekstil. Hasil penelitian diharapkan dapat bermanfaat
sebagai metode alternatif pengolahan limbah tekstil yang dapat digunakan oleh
berbagai stakeholder.
Penelitian dilakukan dalam empat tahapan. Tahapan pertama adalah
karakterisasi DSE berdasarkan kemampuan dekolorisasi zat warna tekstil yaitu
RBBR (antraquinon) dan AB 113 (azo) serta penyisihan logam Cr(VI). Pada tahap
ini digunakan enam cendawan DSE yaitu KSP, PP, CPP, TKC, K.III.3.4, dan DD.
Kemampuan dekolorisasi zat warna dievaluasi dalam media padat dan cair yang
masing-masing mengandung 50 mg/L zat warna RBBR dan AB 113. Begitupun
dengan kemampuan penyisihan Cr(VI) dievaluasi pada media padat dan cair yang
mengandung berbagai konsentrasi Cr(VI), yaitu 10, 30 dan 50 mg/L. Kemampuan
dekolorisasi dan penyisihan Cr(VI) dihitung berturut-turut berdasarkan efisiensi
dekolorisasi (%) dan efisiensi penyisihan (%).
Penelitian tahap kedua adalah uji efisiensi dekolorisasi pada berbagai
kondisi lingkungan yaitu variasi konsentrasi zat warna (100, 200, 250, dan 300
mg/L), pH (3, 5, 7, 9, dan 11) dan salinitas (1, 3, 5, dan 7%). Cendawan yang
digunakan dalam tahap ini adalah tiga cendawan DSE yang menghasilkan
efisiensi dekolorisasi tertinggi pada tahap pertama yaitu strain KSP, PP dan CPP.
Penelitian tahap ketiga adalah pengolahan limbah tekstil menggunakan
cendawan DSE dengan tiga pendekatan yaitu sistem kultur tunggal tanpa
imobilisasi, kultur tunggal terimobilisasi pada Light Expanded Clay Aggregate
(LECA) dan konsorsium tanpa imobilisasi. Pada tahap ini digunakan dua
cendawan yang menghasilkan efisiensi dekolorisasi tertinggi pada tahap dua (KSP
dan PP) dan satu cendawan yang menghasilkan penyisihan Cr(VI) tertinggi kedua
pada tahap satu (TKC). Limbah yang digunakan adalah limbah tekstil dari PT.
Coats Indonesia.
Tahap keempat adalah pengolahan limbah tekstil berbasis fitoremediasi dan
mikoremediasi melalui integrasi sistem FTW menggunakan mangrove
(Rhizophora sp.) dan augmentasi tanaman mangrove dengan DSE pada sistem
terbaik pada hasil tahap ketiga yaitu kultur tunggal strain KSP tanpa imobilisasi.
Percobaan dilakukan dalam skala semi lapangan dengan sistem batch. Pengukuran
kualitas air limbah tekstil hasil pengolahan tahap tiga dan empat dianalisis
berdasarkan karakteristik fisika kimia dan nutriennya. Frekuensi kolonisasi (%)
dihitung untuk menggambarkan keberhasilan kolonisasi DSE pada akar mangrove.
Semua cendawan DSE yang diuji coba mampu mendekolorisasi zat warna
tekstil RBBR dan AB 113 pada konsentrasi 50 mg/L serta menyisihkan logam
berat Cr(VI) dengan konsentrasi 10-50 mg/L. Tiga strain DSE mempunyai
efisiensi dekolorisasi tertinggi untuk zat warna RBBR dan AB 113 yaitu KSP
(88,87% dan 96,99%), PP (81,43% dan 94,34%) dan CPP (73,53% dan 93,81%).
Tiga strain DSE dengan efisiensi penyisihan Cr(VI) tertinggi yaitu PP (99,75–
99,92%), TKC (99,75–99,88%) dan KSP (98,86–99,66%). Berdasarkan hasil
tahap ini, tiga strain DSE yaitu KSP, PP dan CPP dipilih untuk penelitian tahap
kedua yaitu efisiensi dekolorisasi pada berbagai kondisi lingkungan.
Efisiensi dekolorisasi cendawan DSE dipengaruhi oleh faktor lingkungan
dan setiap cendawan DSE mempunyai kondisi optimum yang berbeda. Kondisi
optimum yang diperlukan KSP untuk mendekolorisasi zat warna RBBR dan AB
113 adalah pH asam (pH 3–5) dengan efisiensi dekolorisasi berturut-turut sebesar
92,73–93,06% dan 98,55–98,77%. Untuk strain CPP, pH optimum untuk
mendekolorisasi zat warna RBBR dan AB 113 berturut-turut pada pH basa (9)
dan pH asam (pH 3-5) dengan efisiensi dekolorisasi berturut-turut sebesar
89,68% dan 90,55–91,94%. Berbeda dengan strain PP, kondisi optimum diperoleh
pada pH netral sampai basa (pH 7–9) dengan efisiensi dekolorisasi sebesar 94,79–
95,04% (RBBR) dan 97,70–96,96% (AB 113). Cendawan DSE mampu bekerja
pada kisaran salinitas 1–7% dengan efisiensi dekolorisasi untuk RBBR dan AB
113 berturut-turut sebesar 61,46–93,28% dan 52,85–96,09%. Dua strain DSE
(KSP dan PP) dengan efisiensi dekolorisasi terbaik pada tahap ini dan satu strain
terbaik kedua (TKC) pada uji penyisihan Cr(VI) digunakan untuk penelitian tahap
ketiga.
Secara umum, semua perlakuan (sistem kultur tunggal tanpa imobilisasi dan
terimobilisasi LECA serta konsorsium tanpa imobilisasi) secara nyata berhasil
mendegradasi limbah tekstil dibandingkan kontrol (P<0,05). Sistem imobilisasi
sel dengan LECA berhasil meningkatkan efisiensi penyisihan fosfat dan ortofosfat
berturut-turut sebesar 90,15–96,27% dan 82,00–86,92%. Konsorsium DSE juga
mampu meningkatkan efisiensi penyisihan kedua nutrien tersebut berturut-turut
sebesar 58,92–73,90% dan 48,49–59,79%. Meskipun demikian, kultur tunggal
KSP tanpa imobilisasi menunjukkan performa terbaik dibandingkan perlakuan
lain, sehingga kultur tunggal KSP digunakan untuk penelitian tahap selanjutnya.

dengan strain KSP lebih meningkatkan efisiensi penyisihan nutrien tersebut
berturut-turut menjadi 67,80%; 78,39%; 90,60%; 93,24%; 97,67%; dan 95,60%.
Lebih jauh, kualitas air yang dihasilkan dari teknologi ini telah memenuhi standar
yang ditetapkan pemerintah Republik Indonesia yang tertuang pada
P.16/Menlhk/setjen/Kum.1/4/2019 tentang tentang baku mutu air limbah.
Cendawan DSE berhasil mengolonisasi akar mangrove dengan frekuensi
kolonisasi sebesar 90%.
Penelitian ini membuktikan bahwa cendawan DSE mempunyai kemampuan
dalam mendekolorisasi zat warna tekstil RBBR dan AB 113 serta menyisihkan
logam berat Cr(VI). Kemampuan dekolorisasi DSE dipengaruhi oleh kondisi
lingkungan seperti konsentrasi zat warna, pH dan salinitas. Cendawan DSE
dengan sistem kultur tunggal tanpa imobilisasi terbukti efektif dalam
mendegradasi limbah tekstil. Kultur tunggal terimobilisasi dalam LECA dan
konsorsium efektif dalam meningkatkan efisiensi penyisihan fosfat dan TP.
Teknologi pengolahan limbah tekstil berbasis fitoremediasi melalui sistem FTW
dengan tanaman mangrove dapat dijadikan sebagai alternatif pengolahan limbah
tekstil dan augmentasi tanaman mangrove dengan cendawan DSE
(mikoremediasi) terbukti lebih meningkatkan performa fitoremediasi. Hasil
penelitian ini diharapkan dapat menyediakan teknologi alternatif pengolahan
limbah tekstil yang pada akhirnya dapat mengurangi pencemaran lingkungan yang
disebabkan limbah tekstil. Hasil ini merupakan laporan pertama pemanfaatan
cendawan DSE dalam berbagai aspek seperti dekolorisasi zat warna tekstil,
penyisihan logam berat Cr(VI) dan remediasi limbah tekstil; imobilisasi cendawan
DSE dalam matrik LECA; pemanfaatan mangrove (Rhizophora sp.) dalam
remediasi limbah tekstil; kolonisasi DSE dalam akar mangrove; dan integrasi

dc.description.abstract | The The textile industry is one of the most important industrial sectors and
ranks first in non-oil and gas exports in Indonesia. This industry has experienced
rapid development and has contributed considerably to the economy and human
welfare. However, the textile industry faces significant challenges, especially in
wastewater treatment. Textile wastewater is a very complex waste containing various
hazardous chemical compounds such as synthetic dyes, heavy metals, and other
organic materials, which have the potential to pollute the environment if released
without being treated first. The two most widely used synthetic dyes in the textile
industry are the anthraquinones and azo groups. These dyes are toxic, carcinogenic,
and harmful to the environment and human health. Therefore, textile wastewater
processing technology is needed to reduce environmental pollution. Treatment of
textile wastewater based on fungi (mycoremediation) or plants (phytoremediation)
has been widely reported. Various fungi and plants have also been widely used as
textile wastewater bioremediation agents. However, until now, there has been no
study on the ability of Dark Septate Endophyte (DSE) fungi and mangrove plants to
remediate textile wastewater, including decolorizing dyes and removing the heavy
metal of chromium (Cr(VI)).
The aims of this study: 1) Characterizing DSE with its potential as a degrader
of synthetic dyes (azo and anthraquinone) and a remover of the heavy metal Cr(VI).
2) Analyzing environmental factors that affect DSE activity in decolorizing synthetic
dyes from the azo and anthraquinone groups. 3) Finding suitable mycoremediation
methods through immobilized cell systems and the DSE consortium to degrade textile
wastewater. 4) Testing integration systems of DSE fungi and Floating Treatment
Wetland (FTW) using mangrove plants (Rhizophora sp.) in degrading textile
wastewater. The research results are expected to be helpful as an alternative method
of processing textile wastewater that various stakeholders can use.
The research was conducted in four stages. The first stage was the
characterization of DSE based on the ability to decolorize textile dyes, namely RBBR
(anthraquinone) and AB 113 (azo), and the removal of the heavy metal Cr(VI)). At
this stage, six DSE fungi were used: KSP, PP, CPP, TKC, K.III.3.4, and DD. The
ability to decolorize dyes and reduce heavy metal Cr(VI) was evaluated in solid and
liquid media containing 50 mg/L of dyes RBBR and AB 113 each. Likewise, the
ability to reduce Cr(VI) was evaluated in solid and liquid media containing various
Cr(VI) concentrations, namely 10, 30, and 50 mg/L. The decolorization and removal
abilities of Cr(VI) were calculated based on decolorization efficiency (%) and
removal efficiency (%), respectively.
The second phase of the research was to test the efficiency of decolorization
under various environmental conditions, namely variations in dye concentration (100,
200, 250, and 300 mg/L), pH (3, 5, 7, 9, and 11), and salinity (1, 3, 5 and 7%). Three
DSE fungi, KSP, PP, and CPP strains, which had the highest decolorization efficiency
in the first stage, were employed in this stage.
The third phase of the research was processing textile wastewater using DSE
fungi with three approaches: a single culture system without immobilization, a single
culture immobilized at Light Expanded Clay Aggregate (LECA), and a consortium
without immobilization. At this stage, two fungi, which produced the highest
decolorization efficiency in stage two (KSP and PP), and one fungus, which produced
the second highest Cr(VI) removal in stage one (TKC), were used. The textile
wastewater from PT. Coats Indonesia was used.
Stage four, namely phytoremediation and mycoremediation-based textile
wastewater treatment through integration of FTW system with mangroves
(Rhizophora sp.) and augmentation of mangrove plants with DSE on the best system
on the results of stage three, namely single culture with KSP strains without
immobilization. The experiment was carried out on a semi-field scale with a batch
system. Measurement of the quality of textile wastewater from the processing of
stages three and four was analyzed based on its physicochemical and nutrient
characteristics. Colonization frequency (%) was calculated to describe the success of
DSE colonization on mangrove roots.
All DSE fungi tested could decolorize RBBR and AB 113 textile dyes at a
concentration of 50 mg/L and remove the heavy metal Cr(VI) to 10–50 mg/L. Three
DSE strains had the highest decolorization efficiency for RBBR and AB 113 dyes,
namely KSP (88.87% and 96.99%), PP (81.43% and 94.34%) and CPP (73.53% and
93%). The three DSE strains with the highest Cr(VI) removal efficiency were PP
(99.75–99.92%), TKC (99.75–99.88%) and KSP (98.86–99.66%). Based on the
results of this stage, three DSE strains, namely KSP, PP, and CPP, were selected for
the second phase of the study, namely decolorization efficiency under various
environmental conditions.
The decolorization efficiency of DSE fungi was influenced by environmental
factors, and each DSE fungus has different optimum conditions. The optimum
conditions for KSP to decolorize RBBR and AB 113 dyes were acidic pH (pH 3–5)
with decolorization efficiencies of 92.73–93.06% and 98.55–98.77%, respectively.
For the CPP strain, the optimum pH for decolorizing RBBR dye is at alkaline pH (9),
while AB 113 is at acidic pH (pH 3-5) with decolorization efficiency of 89.67% and
90.55–91.94% respectively. In contrast to the PP strain, optimum conditions were
obtained at neutral to alkaline pH (pH 7–9) with decolorization efficiencies of 94.79–
95.04% (RBBR) and 97.70–96.96% (AB 113). DSE fungi could work in the 1–7%
salinity range with decolorization efficiencies for RBBR and AB 113 of 61.46–
93.28% and 52.85–96.09%, respectively. Two DSE strains (KSP and PP) with the
best decolorization efficiency at this stage and one second-best strain (TKC) in the
Cr(VI) removal test were used for the third phase.
In general, all treatments (single culture system without immobilized and
immobilized LECA and consortium without immobilization) significantly succeeded
in degrading textile waste compared to controls (P<0.05). The cell immobilization
system with LECA increased the phosphate and orthophosphate removal efficiency
by 90.15–96.27% and 82.00–86.92%, respectively. The DSE consortium also
increased the removal efficiency of the two nutrients by 58.92–73.90% and 48.49–
59.79%, respectively. Nevertheless, KSP single culture without immobilization
performed better than other treatments, so KSP single culture was used for further
research.
Phytoremediation-based textile wastewater treatment through the FTW system
with mangrove plants (Rhizophora sp.) showed removal efficiency in color, N-NH4

92.32%, respectively. Mangrove augmentation with KSP strains further increased the
removal efficiency of these nutrients to 67.80%, 78.39%, 90.60%, 93.24%, 97.67%,
and 95.60%, respectively. Furthermore, the quality of water produced from this
technology meets the standards set by the government of the Republic of Indonesia,
which are contained in P.16/Menlhk/setjen/Kum.1/4/2019 concerning wastewater
quality standards. In addition, DSE fungi have succeeded in colonizing mangrove
roots with a colonization frequency of 90%
This study proved that the DSE fungi can decolorize RBBR and AB 113 textile
dyes and remove the heavy metal Cr(VI). Environmental conditions such as dye
concentration, pH, and salinity affect DSE's decolorization ability. DSE fungi with a
single culture system without immobilization proved effective in degrading textile
wastewater. Immobilized single cultures in LECA and consortium effectively
increased phosphate and TP removal efficiency. Textile wastewater treatment
technology based on phytoremediation through the FTW system with mangrove
plants can be used as an alternative to textile wastewater processing, and the
augmentation of mangrove plants with DSE fungi (mycoremediation) was proven to
improve the performance of phytoremediation further. The results of this study were
expected to provide alternative technologies for processing textile wastewater, which
in turn can reduce environmental pollution caused by textile wastewater. This result
is the first report regarding the use of DSE fungi in various aspects such as
decolorization of textile dyes, removal of heavy metal Cr(VI) and remediation of
textile wastewater; immobilization of DSE fungi in the LECA matrix; utilization
of mangroves (Rhizophora sp.) in remediating textile wastewater; DSE
colonization success in mangrove roots; and integration of FTW systems and DSE
fungi augmentation in textile wastewater treatment. | id |
