Optimasi Formula Enkapsulasi Ekstrak Propolis Trigona Menggunakan Maltodekstrin-Gum Biji Selasih dengan Teknik Spray Drying
Abstract
Gum biji selasih merupakan salah satu alternatif sumber gum alami yang berasal dari bahan baku lokal. Gum biji selasih memiliki kemampuan membentuk film yang berpotensi digunakan sebagai bahan penyalut dalam proses enkapsulasi. Salah satu bahan yang berpotensi untuk dienkapsulasi ialah propolis karena memiliki senyawa fenolik yang bersifat sebagai antioksidan, antimikroba, dan antikanker. Penerapan propolis sebagai ingridien pangan fungsional memiliki keterbatasan, antara lain kelarutan yang rendah dan terdegradasinya senyawa bioaktif pada suhu tinggi. Enkapsulasi dapat menjadi solusi efektif dalam mengatasi keterbatasan tersebut. Penggunaan kombinasi maltodekstrin dan gum biji selasih sebagai bahan penyalut enkapsulasi ekstrak propolis belum pernah dilakukan. Tujuan penelitian ini adalah menghasilkan formula optimum serbuk enkapsulat ekstrak propolis dengan menggunakan kombinasi bahan penyalut (maltodekstrin dan gum biji selasih) dengan teknik spray drying, serta menentukan karakteristik fisikokimia serbuk enkapsulat ekstrak propolis optimum yang dihasilkan.
Penelitian ini terdiri dari 4 tahapan. Tahap pertama, yakni persiapan dan karakterisasi bahan baku (gum kasar biji selasih kering dan ekstrak propolis). Tahap kedua, yakni optimasi formula enkapsulasi ekstrak propolis dengan teknik spray drying menggunakan response surface methodology (RSM). Tahap ketiga, yakni verifikasi formula optimum enkapsulat ekstrak propolis, dan tahap keempat yakni karakterisasi serbuk enkapsulat ekstrak propolis terpilih. Variabel bebas yang digunakan untuk optimasi pada tahap kedua, yakni 1–10 g ekstrak propolis cair (faktor 1) dan 0–0,5 gum kasar biji selasih kering (faktor 2). Maltodekstrin sebanyak 10 g dan akuades sebanyak 100 mL ditetapkan sebagai variabel tetap. Respon yang diukur yaitu, kadar air, kelarutan, dan total fenol.
Kadar total fenol gum kasar biji selasih kering dan ekstrak propolis diperoleh masing-masing 1866 and 1461 mg ekuivalen asam galat (EAG)/kg sampel (basis basah (bb)). Formula optimum terpilih dari hasil optimasi dengan RSM berdasarkan keseluruhan respon, yakni 10 g propolis; 0,5 g gum kasar biji selasih kering; 10 g maltodekstrin; dan 100 mL akuades dengan nilai desirability sebesar 0,734. Serbuk enkapsulat ekstrak propolis optimal memiliki kadar air 3,82% (bb), kelarutan 96,13% (basis kering), total fenol 901,84 mg EAG/kg sampel (bb), aktivitas air 0,41, higroskopisitas 16.82% (bb), dan kadar lemak 4,61% (bb). Hasil analisis aktivitas antioksidan serbuk enkapsulat optimum dengan metode DPPH (2,2-diphenyl-1-picrylhydrazyl) mendapatkan persentase inhibisi sebesar 17,61% (bb) pada konsentrasi 5000 µg/mL dan kapasitas antioksidan sebesar 2090 mg ekuivalen asam askorbat (EAA)/kg sampel (bb). Sedangkan aktivitas antioksidan serbuk enkapsulat optimum dengan metode FRAP (ferric reducing antioxidant power) mendapatkan kapasitas antioksidan sebesar 2394 µmol EAA/kg sampel (bb). Pengamatan morfologi serbuk enkapsulat ekstrak propolis optimal menunjukkan bentuk partikel partikel yang teraglomerasi. Basil seed gum is an alternative source of natural gum based on local product. Basil seed gum has the film forming ability that have the potential to be used as a wall material for encapsulation. One of the materials that has the potential to be encapsulated is propolis because it has phenolic compounds that act as antioxidant, antimicrobial, and anticancer. The application of propolis as a functional food ingredient has some limitations, including low solubility and the degradation of bioactive compounds at high temperatures. Encapsulation can be an effective solution to overcome these limitations. The use of a combination of maltodextrin and basil seed gum as a coating material for the encapsulation of propolis extract has never been done. The objectives of this research were to produce encapsulated
propolis extract powders in optimum formula using a combination of coating materials (maltodextrin and basil seed gum) by spray drying technique, and to determine the physicochemical characteristics of the resulting optimum product.
This research consists of 4 stages. The first stage is the preparation and characterization of raw materials (dried crude basil seed gum and propolis extract). The second stage is optimizing the propolis extract encapsulation formula by spray drying technique using the response surface methodology (RSM). The third stage is the verification of the optimum formula for encapsulated propolis extract powders, and the fourth stage is the characterization of the selected encapsulated propolis extract powders. The independent variables used for optimization in the second stage were 1–10 g of liquid propolis extract (factor 1) and 0–0.5 g of dried crude basil seed gum (factor 2). 10 g of maltodextrin and 100 mL of aquades were set as fixed variables. The responses measured were moisture content, solubility, and total phenolic content (TPC).
The total phenolic of dried crude basil seed gum and propolis extract were 1866 and 1461 mg gallic acid equivalent (GAE)/kg sample (wet basis (wb)), respectively. The resulting optimum formula from RSM based on the overall response was 10 g of liquid propolis extract, 0.5 g of dried crude basil seed gum,
10 g of maltodextrin, and 100 mL of aquades with desirability value of 0.734. The optimum encapsulated propolis extract powders had moisture content of 3.82% (wb), solubility of 96.13% (dry basis), and TPC of 901.84 mg GAE/kg sample (wb), water activity value of 0.41, hygroscopicity value of 16.82% (wb), and fat content of 4.61% (wb). The results of the analysis of the antioxidant activity of the optimum product by the DPPH (2.2-diphenyl-1- picrylhydrazyl) assay had an inhibition percentage of 17.61% (wb) at a concentration of 5000 µg/mL and an antioxidant capacity of 2091 mg ascorbic acid equivalent (AAE)/kg sample (wb). While antioxidant activity of the optimum product by the FRAP (ferric reducing antioxidant power) assay gave an antioxidant capacity of 2394 µmol AAE/kg sample (wb). Morphological observations of the selected encapsulated propolis extract powders showed the formation of agglomerated particles.
