Identifikasi Mikroflora pada Produk Fermentasi Ikan Pado

Ikan pado merupakan salah satu produk fermentasi indigenous Sumatera Barat yang cukup unik dan dibuat dari ikan laut segar berukuran kecil yang sebelumnya telah dibersihkan dari insang, sirip dan isi perut, selanjutnya difermentasikan dengan campuran ampas kelapa dan cacahan daging biji picung (Pangium edule Reinw) kering. Proses fermentasi dilakukan di dalam wadah tertutup yang dilapisi plastik selama 5-7 hari pada suhu ruang (25-30°C). Penggunaan daging biji picung kering dalam proses fermentasi diyakini memiliki peranan penting dalam meningkatkan keawetan alami produk. Ikan pado merupakan produk fermentasi yang berbentuk ikan utuh, akan tetapi berwarna coklat kehitaman, agak kering serta mempunyai rasa dan aroma yang khas. Identifikasi mikroflora pada produk ikan pado belum pernah dilaporkan, baik itu pada produk akhir maupun perubahan populasi mikroflora selama proses fermentasi. Oleh sebab itu, penelitian ini bertujuan untuk mengidentifikasi dan mempelajari perubahan populasi mikroflora serta karakteristik selama proses fermentasi, mengidentifikasi mikroflora dan karakteristik ikan pado yang dipasarkan serta mengidentifikasi mikroorganisme dominan pada produk secara molekuler. Hasil penelitian menunjukkan bahwa selama proses fermentasi terjadi peningkatan populasi bakteri asam laktat dari 3.48 log CFU/g menjadi 8.45 log CFU/g dan angka lempeng total dari 4.25 log CFU/g menjadi 7.64 log CFU/g. Sementara itu, total Enterobacteriaceae mengalami penurunan dari 0.50 log CFU/g menjadi <1 log CFU/g dan total pembentuk spora dari 0.94 log CFU/g menjadi <1 log CFU/g. Angka lempeng total, total bakteri asam laktat, angka kapang khamir dan total Enterobacteriaceae ikan pado sampel pasar (Produk A dan B) relatif sama-, akan tetapi angka pembentuk spora pada kedua produk berbeda nyata yakni <1 log CFU/g dan >5 log log CFU/g. Proses fermentasi juga menurunkan kadar air produk ikan pado. Sebanyak 77 isolat diperoleh dari ikan pado dalam penelitian ini, diantaranya 36 isolat bakteri asam laktat, dan 17 isolat bakteri pembentuk spora yang sebagai Bacillus sp, dan 24 isolat kapang yang merupakan golongan Penicillium sp, Rhizopus sp, Aspergillus sp dan Geotricum sp.
Identifikasi menggunakan gen penyandi 16S rRNA dilakukan pada bakteri yang dominan dalam ikan pado yakni 14 dari 36 isolat bakteri asam laktat yang terdiri dari 10 isolat yang didapatkan selama proses fermentasi dan 4 isolat dari sampel pasar. Seluruh isolat yang diidentifikasi memiliki kekerabatan terdekat dengan Lactiplantibacillus plantarum dengan nilai hit yang tinggi dan juga Lactiplantibacillus pentosus dengan nilai hit rendah. Pado fish is one of typical and unique fermented products from West Sumatra. Pado fish is made from small pelagic fish mixed with dried picung (Pangium edule Reinw) seed and grated coconut and stacked up in a closed container for five to seven days at room temperature. The use of dried Pangium edule during fermentation process has been assumed to be the important factors that enhance the shelf-life of the product. The fermented pado fish visually remains as whole fish with blackish brown color, slightly dry, brittle and has unique taste and aroma. Microflora on pado fish has never been documented, either in the final product or the changes of microflora population during the fermentation. Therefore, the aims of this study were to determine the microflora during fermentation and from the pado fish avalaible in the market and to identify the predominant microorganisms using molecular approaches.
The results showed that during fermentation there was an increase in the population of lactic acid bacteria from 3.48 log CFU/g to 8.45 log CFU/g and and total plate count from 4.25 log CFU/g to 7.64 log CFU/g, repectively. Meanwhile the total Enterobacteriaceae decreased from 0.50 log CFU/g to <1 log CFU g and spore forming bacteria from 0.94 log CFU/g to <1 log CFU/g, respectively. The population of microflora in pado from market (Product A and B) had relatively similar total plate count, lactic acid bacteria count, yeast and mould number, and total enterobacteriaceae count. However, they showed significantly different numbers of spore forming bacteria, which is <1 log CFU/g in produk A and > 5 log CFU/g in product B. Fermentation process also reduce the moisture content, pH and aw of the products. Seventy-seven isolates were obtained from the pado fish in this reseach, thirty-six isolate were lactic acid bacteria, seventeen isolates were spore forming bacteria as Bacillus sp and twenty four mold isolates to be Penicillium sp, Rhizopus sp, Aspergillus sp dan Geotricum sp.
Identification using 16S rRNA coding gene were carried out on predominant bacteria in pado fish, that was fourteen of thirty-six isolates of lactic bacteria, including ten isolates obtained during fermentation process and four isolates from the market samples. Almost all isolates showed the closest relationship with Lactiplantibacillus plantarum with high number of hits and with Lactiplantibacillus pentosus with low number of hits.
