Sperm plasma membrane integrity was important for sperm fertility. The objective of this study was to determine the optimal time to test sperm plasma membrane of raw boar semen using hypo osmotic swelling (HOS) test. A total of 10 sexualy mature boars from three breed (landrace, Duroc, and Yorkshire) used as a sperm source. Semen were collected using hand glove method . Immediatley after collection the semen were evaluate macro and microscopica lly. The HOS test conducted by putting 50 j..tL semen into 1 rnl HOS medium (150 mOsm Kg-1 incubated at 37°C. Two hundred reacted and not reacted sperm cell to HOS medium were evaluate at 0 min and every 15 minutes. Result showed that theoptimum response to HOS was obtained at 60 minutes after incubation.id
