| dc.description.abstract | The effect of glucose concentration on in vitro development of preimplantation mouse embryos was investigated using M16 culture medium supplemented with 3% BSA. Mouse embryos step of one cell from CBR were cultured in M16-BSA culture medium containing glucose with concentrations of (1) 0 mM; (2) 2.5 mM; (3) 5.0 mM; (4) 7.5 mM and (5) 10.0 mM, successively without 1.19 mM phosphate. The results showed that glucose did not inhibit the development of CBR preimplantation mouse embryos, but glucose concentration did influence the development rate. After 24 hrs cultured, supplementation of glucose showed beneficial for development of 1-cell to 2- cell embryos (93.8 - 100 8, p<0,05), compared to one without (3 1.6%). Beyond 8-cell stage, the concentration of 2.5 mM glucose showed a high percentage of 1-cell embryos that developed to 4-cell stage (82.0%, p<0,05) compared to the concentration of 5.0 mM (46.7%), 7,5 mM (28.9%) and 10.0 mM (27.9%), respectively. In medium containing 2.5 mM glucose, 8-cell embryos that developed to CM-stages decreased (33.3%) and none developed to the blastocyst (BL) stage. Compared to one containing 5.0 mM glucose, embryos that developed to the CM- and BLstages were (66.7%) and (50.0%), while concentration of 10.0 mM were (75.0%) and (66.7%), respectively. Addition of 1.19 mM phosphate in medium increased the percentage of embryos developed to morula and blastocyst when glucose was presence in low concentration (2.5 mM) compared to one without.
