Tempe merupakan pangan khas Indonesia yang memiliki aktivitas hipoglikemik, yaitu aktivitas yang berkaitan dengan kemampuan untuk mengontrol kadar glukosa darah. Aktivitas ini terkait dengan penyakit diabetes. Tempe memiliki beberapa dugaan mekanisme hipoglikemik yang bekerja di pankreas dan usus halus. Senyawa dalam tempe yang diduga mampu memperbaiki fungsi pankreas adalah senyawa antioksidan dan asam amino bebas insulinotropik, sedangkan senyawa inhibitor α-glukosidase yang bekerja pada usus halus dapat menghambat penyerapan glukosa.
Tempe biasa dikonsumsi oleh masyarakat secara segar tepat setelah dua hari fermentasi. Setelah lewat satu atau dua hari, masyarakat enggan mengonsumsinya karena mutunya dianggap menurun dan tidak sebaik tempe segar. Padahal mutu gizi dan hipoglikemiknya dilaporkan mengalami peningkatan jika waktu fermentasinya diperpanjang. Selain itu, mutu tempe dapat ditingkatkan dengan menggunakan kedelai germinasi sebagai bahan bakunya. Kedelai germinasi memiliki mutu gizi dan hipoglikemik yang lebih baik dari kedelai biasa atau non-germinasi. Berdasarkan hal tersebut, penggunaan kedelai germinasi sebagai bahan baku tempe dapat dikombinasikan dengan penambahan waktu fermentasi guna meningkatkan mutu tempe yang dihasilkan.
Penelitian ini bertujuan untuk mengevaluasi karakteristik fisikokimia tempe dan potensinya sebagai pangan hipoglikemik pada beberapa mekanisme akibat penggunaan kedelai germinasi dan penambahan waktu fermentasi. Terdapat dua perlakuan dalam penelitian ini, yaitu waktu fermentasi tempe (48, 72, dan 96 jam) dan bentuk kedelai yang digunakan (kedelai germinasi dan kedelai non-germinasi yang biasa ditemukan dalam pembuatan tempe). Analisis yang dilakukan meliputi analisis fisikokimia (warna, tekstur, sensori, proksimat, aktivitas air, pH, total asam tertitrasi, protein terlarut, profil peptida, profil asam amino bebas) dan analisis potensi hipoglikemik (asam amino bebas insulinotropik, isoflavon aglikon, total senyawa fenolik, aktivitas antioksidan, dan aktivitas inhibitor α-glukosidase).
Secara fisik, tempe mengalami penurunan kekerasan akibat penggunaan kedelai germinasi dan penambahan waktu fermentasi 48 hingga 96 jam, terlihat dari pengukuran menggunakan texture analyzer dan pengamatan sensori. Hasil uji hedonik terhadap warna tempe juga dilaporkan menurun akibat kedua perlakuan, walaupun tidak berbeda signifikan menurut hasil pengukuran dengan chromameter. Hasil uji hedonik untuk kesukaan keseluruhan mengalami penurunan, namun masih dapat diterima. Komposisi proksimat dan aktivitas air tempe tidak mengalami perubahan akibat penggunaan kedelai germinasi dan penambahan waktu fermentasi, kecuali kadar air. Perlakuan germinasi dapat meningkatkan kadar air tempe yang difermentasi 48 hingga 96 jam akibat matriks kedelai germinasi dan penambahan air selama proses germinasi dari penyiraman berkala.
Nilai pH, kadar protein terlarut, dan asam amino bebas tempe mengalami peningkatan seiring dengan penambahan waktu fermentasi. Pada waktu fermentasi 72 dan 96 jam, tempe kedelai non-germinasi memiliki kadar protein terlarut, asam amino bebas, dan peptida yang lebih tinggi dari tempe kedelai germinasi. Hal ini menguatkan dugaan adanya hidrolisis protein yang lebih intens di tempe kedelai non-germinasi pada waktu fermentasi tambahan. Konsentrasi peptida yang relatif tinggi yang ditemukan dalam penelitian ini seiring dengan penambahan waktu fermentasi tempe, membuka peluang untuk penelitian eksplorasi peptida bioaktif.
Pengamatan pada potensi hipoglikemik menunjukkan penambahan waktu fermentasi dari 48 hingga 96 jam dapat meningkatkan potensi hipoglikemik tempe pada mekanisme perbaikan fungsi pankreas, dilihat dari kadar asam amino bebas insulinotropik dan senyawa antioksidan. Kedelai germinasi yang difermentasi 96 jam menghasilkan tempe yang tinggi asam amino bebas insulinotropik, sedangkan pada waktu fermentasi yang sama, kedelai non-germinasi lebih unggul dalam hal parameter antioksidan, seperti isoflavon, total senyawa fenolik, dan kapasitas antioksidan. Pada mekanisme senyawa inhibitor α-glukosidase, potensi hipoglikemik tempe akibat perlakuan germinasi dan penambahan waktu fermentasi mengalami penurunan, dilihat dari nilai aktivitasnya. Terdapat dugaan bahwa hal ini terkait dengan penurunan peptida berukuran 11,40 kDa yang dominan pada tempe dalam studi ini.
Dari hasil penelitian ini, tempe yang difermentasi hingga 96 jam memiliki potensi hipoglikemik lebih unggul pada mekanisme perbaikan pankreas daripada tempe yang difermentasi 48 jam, walaupun mutu fisiknya mengalami penurunan. Dengan waktu fermentasi 96 jam, tempe dari kedelai germinasi lebih unggul dengan tingginya asam amino bebas insulinotropik, sedangkan tempe dari kedelai non-germinasi lebih unggul dengan tingginya komponen antioksidannya. Tempe kedelai non-germinasi yang difermentasi 48 jam juga memiliki keunggulan potensi hipoglikemik dilihat dari aktivitas inhibitor α-glukosidasenya, selain keunggulan mutu fisik yang dimilikinya. Berdasarkan hasil ini, tempe yang difermentasi 96 jam baik dari kedelai germinasi maupun non-germinasi dapat dikonsumsi oleh penderita diabetes sebagai upaya penyembuhan, sedangkan tempe yang difermentasi 48 jam dapat dikonsumsi oleh orang normal untuk upaya pencegahan terhadap penyakit diabetes. Tempe is a typical Indonesian food which has hypoglycemic activity. This activity is related to the ability to control blood glucose levels associated with diabetes. Tempe has several suspected hypoglycemic mechanisms which work in pancreas and small intestine. The suspected compounds to improve pancreatic function are antioxidant compounds and insulinotropic free amino acids, while α-glucosidase inhibitor compounds in tempe can inhibit glucose absorption in small intestine.
Tempe is usually consumed in fresh form by the people right after two days fermentation. After one or two days, people are reluctant to consume it, because of its decrease quality and it is not as good as fresh tempe. In fact, the nutritional and hypoglycemic quality is reported to have increased if the fermentation time was extended. In addition, the quality of tempe can be improved by using germinated soybeans as the raw material. Germinated soybeans have better nutritional and hypoglycemic qualities than regular or non-germinated soybeans. Based on this, the use of germinated soybeans as raw material for tempe can be combined with the addition of fermentation time to improve the quality of the tempe produced.
This study aimed to evaluate the physicochemical characteristics of tempe and its potential as a hypoglycemic food in several mechanisms due to the use of germinated soybeans and extended fermentation time. Two treatments applied in this study were tempe fermentation time (48, 72, and 96 hours) and the form of soybean used (germinated soybean and non-germinated which is usually found in tempe production). The analysis included physicochemical analysis (color, texture, sensory, proximate, water activity, pH, total titrated acid, soluble protein, peptide profile, free amino acid profile) and analysis of hypoglycemic potential (insulinotropic free amino acids, isoflavones. aglycones, total phenolic compounds, antioxidant activity, and α-glucosidase inhibitor activity).
Physically, tempe experienced a decrease in texture due to the use of germinated soybeans and an additional fermentation time of 48 to 96 hours, considered from the observation using a texture analyzer and sensory examination. The result of tempe hedonic color was also decreased due to two treatments, although it did not significantly changed according to the chromameter measurement. The overall hedonic score had decreased, but was still acceptable. The proximate composition and water activity of tempe did not change due to the use of germinated soybeans and the addition of fermentation time, except for the water content. The germination treatment increase the water content of fermented tempe for 48 to 96 hours due to the matrix of germinated soybean and the water absorption during germination process periodic watering.
The pH value, soluble protein content, and free amino acids of tempe increased with the addition of fermentation time. At the fermentation time of 72 and 96 hours, non-germinated soybean tempe had higher levels of soluble protein, free amino acids, and peptides than germinated soybean tempe. This confirmed the suspicion of a more intense protein hydrolysis in non-germinated soybean tempe during extended fermentation time. The high number of peptides concentration found in this study with extended tempe fermentation opened the opportunity for exploration research of bioactive peptides.
Observations on the hypoglycemic potential showed that increasing the fermentation time from 48 to 96 hours increase the hypoglycemic potential of tempe in the mechanism of repairing pancreatic function, showed from the levels of insulinotropic free amino acids and antioxidant compounds. Germinated soybeans fermented for 96 hours produced tempe which was high in insulinotropic free amino acids, while at the same fermentation time, non-germinated soybeans were superior in terms of antioxidant parameters, such as isoflavones, total phenolic compounds, and antioxidant capacity. In the mechanism of α-glucosidase inhibitor compounds, the hypoglycemic potential of tempe due to germination treatment and the extended fermentation time has decreased, showed from the value of its activity. This could be related to the decrease in the dominant peptide size of 11.40 kDa in tempe in this study.
From the results of this study, tempe that fermented for up to 96 hours had a superior hypoglycemic potential on improving pancreatic function compared to tempe fermented for 48 hours, although its physical quality had decreased. With a fermentation time of 96 hours, tempe from germinated soybeans was superior with high levels of insulinotropic free amino acids, while tempe from non-germinated soybeans was superior with high antioxidant components. Non-germinated soybean tempe fermented for 48 hours also has the advantage of hypoglycemic potential seen from its α-glucosidase inhibitor activity, while it had superior physical quality. Based on these results, fermented soybean 96 hours from both germinated and non-germinated soybeans can be consumed by diabetic patients as an effort to cure, while fermented tempe for 48 hours can be consumed by normal people for prevention of diabetes.
